Eladon is a bronchodilating, anti-inflammatory and H1-antihistamine drug used to treat respiratory diseases.

Release form and composition

Dosage forms of Eladon:

  • prolonged-release film-coated tablets: biconvex, round; the core in a cross section and the shell - almost white or white (10, 15 or 30 pieces in a blister strip; in a cardboard box 3, 6 or 9 packs of 10 pieces; 2, 4 or 6 packs of 15 pieces; 1, 2 or 3 packs of 30 pcs.; 30 pcs. In a polyethylene can, in a cardboard box 1 can);
  • syrup: clear or slightly opalescent, brown-yellow liquid; during storage, the formation of a precipitate is allowed, which disappears when shaken (150 or 250 ml in dark-colored polyethylene terephthalate bottles; in a cardboard box 1 bottle with or without a dispensing spoon).

1 tablet contains:

  • active substance: fenspiride hydrochloride - 80 mg;
  • additional components: calcium hydrogen phosphate dihydrate, collidone SR (silicon dioxide 0.2%, sodium lauryl sulfate 0.8%, povidone 19%, polyvinyl acetate 80%), magnesium stearate, colloidal silicon dioxide;
  • film shell: hyprolose (hydroxypropyl cellulose), hypromellose, talc, titanium dioxide.

1 ml of syrup contains:

  • active substance: fenspiride hydrochloride - 2 mg;
  • additional components: glycerol (glycerin), propyl parahydroxybenzoate (propylparaben), methyl parahydroxybenzoate (methylparaben), sodium saccharinate, sucrose, potassium sorbate, dried licorice root extract, purified water, food flavoring "Med-96", flavoring "Vanilla15460", food mint flavoring.

Pharmacological properties

Pharmacodynamics

Fenspiride, the active ingredient of Eladon, exhibits anti-inflammatory and anti-bronchoconstrictor effect due to a decrease in the production of a number of biologically active substances [derivatives of arachidonic acid, cytokines, mainly tumor necrosis factor α (TNF-α), free radicals], which play an important role in the occurrence of bronchospasm and inflammation.

The suppression of the metabolism of arachidonic acid by the substance is enhanced by its H1-antihistaminic activity, since histamine accelerates the metabolic transformation of arachidonic acid with the formation of prostaglandins and leukotrienes. The agent reduces the secretion of the bronchial glands, ensuring the blocking of α-adrenergic receptors, the stimulation of which is accompanied by increased secretion production.

Thus, fenspiride leads to a weakening of the influence of a number of factors that contribute to the hypersecretion of pro-inflammatory agents, the development of the inflammatory process and bronchial obstruction. The active substance also has antispasmodic properties.

Pharmacokinetics

After oral administration, the drug is well absorbed from the gastrointestinal tract. The maximum concentration (Cmax) of fenspiride in blood plasma is achieved after 6 hours (for tablets) or 2.3 ± 2.5 hours (for syrup). The half-life (T1 / 2) of the substance is 12 hours, 90% of the drug is excreted by the kidneys and 10% through the intestines.

Indications for use

According to the instructions, Eladon is recommended for the treatment of the following diseases of the upper and lower respiratory tract:

  • bronchial asthma (as part of combination therapy);
  • bronchitis (with or without chronic respiratory failure);
  • tracheobronchitis;
  • laryngitis and rhinopharyngitis;
  • sore throat, cough, hoarseness against the background of flu, whooping cough and measles;
  • infectious lesions of the respiratory tract, proceeding with cough, in cases where standard antibiotic therapy is prescribed.

Also, the drug is used to treat otitis media and sinusitis of various genesis, including allergic.

Contraindications

Absolute:

  • age up to 2 years - for syrup, up to 18 years - for tablets;
  • pregnancy and the period of breastfeeding;
  • glucose-galactose malabsorption, fructose intolerance, sucrase / isomaltase deficiency - for syrup;
  • hypersensitivity to the components of the drug.

Relative (it is recommended to take Eladon with caution):

  • simultaneous reception with sedatives, ethanol;
  • diabetes mellitus - for syrup (due to the presence of sucrose in the composition).

Instructions for the use of Eladon: method and dosage

Eladon is taken orally. The duration of treatment is established by the attending physician.

Sustained-release film-coated tablets

Adults should take 80 mg 2-3 times a day (1 piece), the maximum daily dose should not exceed 240 mg (3 Eladon tablets).

Syrup

Children over 2 years of age should take 4 mg / kg syrup per day.

Recommended daily doses:

  • body weight less than 10 kg: 2–4 tsp. (10–20 ml), syrup can be poured into a feeding bottle;
  • body weight over 10 kg: 2-4 tbsp. l. (30-60 ml) before meals.

For adolescents and adults, Eladon is prescribed 3–6 tablespoons (45–90 ml) per day, before meals.

Shake well before taking. 5 ml (1 tsp.) And 15 ml (1 tbsp. L.) Syrup contains 10 and 30 mg fenspiride hydrochloride, respectively.

Side effects

  • nervous system: rarely - drowsiness; with an unknown frequency - dizziness;
  • digestive system: often - pain in the epigastric region, nausea, gastrointestinal upset; with an unknown frequency - vomiting, diarrhea;
  • cardiovascular system: rarely - moderate tachycardia, the severity of which decreases with a decrease in the dose of the drug; with an unknown frequency - a decrease in blood pressure, palpitations, probably due to tachycardia;

Overdose

If you suspect an overdose of the drug (cases of overdose have been recorded when using the drug in a dose over 2320 mg), you should immediately consult a doctor.

Overdose signs may include agitation / drowsiness, sinus tachycardia, vomiting, nausea. In this condition, gastric lavage, symptomatic therapy, monitoring of the electrocardiogram, measures aimed at maintaining the vital functions of the body are prescribed.

With the development of infections of the respiratory system, excited by fungi or bacteria, Eladon is recommended to be taken only in combination with antibacterial and antifungal agents.

The syrup contains parabens (parahydroxybenzoates), as a result of which the intake of this drug can lead to the development of allergic reactions, including delayed ones.

Patients with diabetes should take into account that 1 tsp. syrup Eladon contains 3 g of sucrose, which approximately corresponds to 0.3 XE, and 1 tbsp. l. - 9 g of sucrose, which approximately corresponds to 0.9 XE.

Influence on the ability to drive vehicles and complex mechanisms

Studies on the effect of fenspiride on the ability to perform work requiring concentration of attention and speed of psychomotor reactions have not been conducted.

Application during pregnancy and lactation

The use of Eladon during pregnancy is contraindicated due to limited data on the use of fenspiride during this period.

Treatment with the drug is not a reason for terminating the onset of pregnancy. There are no clinical data confirming the fetotoxic effect of fenspiride or its ability to cause malformations during the course of therapy during pregnancy.

Since there is no information about the penetration of fenspiride into breast milk, the drug should not be taken during lactation. If it is necessary to use the drug during this period, it is necessary to resolve the issue of stopping breastfeeding.

Pediatric use

Syrup is contraindicated for children under 2 years old, Eladon tablets - under 18 years old.
