Ezlor is a drug with antiallergic effect.

Release form and composition

Ezlor's dosage form is film-coated tablets: cylindrical biconvex, the core and shell are white or almost white (in a cardboard box 1-6, 8 or 10 blisters of 10 tablets or 1 polymer can of 10, 20, 30, 40, 50 or 100 tablets).

Composition of 1 tablet:

  • active substance: desloratadine - 5 mg;
  • auxiliary components: lactose monohydrate - 70 mg; microcrystalline cellulose - 12.1 mg; povidone - 3 mg; magnesium stearate - 0.9 mg; croscarmellose sodium - 4 mg;
  • shell: white Opadry II 85F48105 - 3 mg (including: polyvinyl alcohol - 1.407 mg; macrogol 3350 - 0.708 mg; talc - 0.522 mg; titanium dioxide - 0.363 mg).

Pharmacological properties

Pharmacodynamics

Desloratadine, the primary active metabolite of loratadine, is a long-acting H 1 histamine receptor blocker.

The therapy allows you to prevent the appearance, and also facilitates the course of allergic reactions. Ezlor has an antipruritic and anti-exudative effect, reduces capillary permeability, prevents smooth muscle spasm and tissue edema.

The drug does not affect the central nervous system, practically does not have a sedative effect (does not cause drowsiness), does not cause prolongation of the QT interval on the ECG, does not change the speed of psychomotor reactions.

Ezlor develops within 30 minutes after oral administration, and lasts 24 hours.

Pharmacokinetics

After oral administration, desloratadine is well absorbed from the gastrointestinal tract. Determined plasma concentrations in the blood are reached within 30 minutes, the time to reach the maximum concentration is approximately 3 hours.

Binds to plasma proteins - in the range from 83 to 87%. When used for 14 days 1 time a day, 5–20 mg in adults and adolescents, there is no clinically significant accumulation of the substance. Simultaneous food intake / consumption of grapefruit juice does not affect the distribution of desloratadine (when taken once a day at a dose of 7.5 mg). The substance does not penetrate through the blood-brain barrier.

Desloratadine undergoes intensive metabolism in the liver by hydroxylation to form 3-OH-desloratadine combined with glucuronide. It is not an inhibitor of CYP3A4 and CYP2D6 isoenzymes, as well as a substrate / inhibitor of P-glycoprotein.

A small amount of the dose taken is excreted unchanged: by the kidneys - less than 2%; through the intestines - less than 7%. The half-life (T 1/2) varies in the range of 20-30 hours (on average - 27 hours).

Indications for use

  • allergic rhinitis: to eliminate / relieve itching in the nose and palate, nasal congestion, sneezing, watery eyes, nasal mucus, redness / itching of the eyes;
  • urticaria: to reduce / eliminate itching, rash.

Contraindications

Absolute:

  • lactose intolerance, lactase deficiency or glucose-galactose malabsorption syndrome;
  • pregnancy and the period of breastfeeding;
  • age up to 12 years;
  • individual intolerance to the components of the drug.

In severe renal failure, Ezlor should be used under medical supervision.

Instructions for the use of Ezlor: method and dosage

Ezlor tablets are taken orally with a small amount of water, preferably at the same time of day. The tablet should not be divided or chewed. The effectiveness of therapy does not depend on the time of the meal.

Recommended dosage regimen: 1 tablet once a day.

Side effects

Possible adverse reactions (> 10% - very common;> 1% and 0.1% and 0.01% and <0.1% - rarely; <0.01% - very rare):

  • central nervous system: infrequently - headache; very rarely - insomnia, hallucinations, convulsions, drowsiness, dizziness, psychomotor hyperactivity;
  • immune system: very rarely - rash, dyspnea, urticaria, angioedema, anaphylactic shock;
  • digestive system: infrequently - xerostomia; very rarely - dyspepsia, nausea, vomiting, diarrhea, increased activity of liver enzymes, abdominal pain, hepatitis, increased concentration of bilirubin;
  • cardiovascular system: very rarely - palpitations, tachycardia;
  • musculoskeletal system: very rarely - myalgia;
  • others: often - increased fatigue.

Overdose

According to clinical and pharmacological studies, taking desloratadine in a daily dose of 45 mg for 10 days did not lead to the development of serious adverse reactions.

Therapy: gastric lavage, intake of activated carbon; if necessary, symptomatic treatment is indicated. Desloratadine is not excreted during hemodialysis, and the effectiveness of peritoneal dialysis has not been determined.
