Dosage form - tablets: white with a cream shade, odorless, bitter in taste (10 pcs. In a blister strip, 2 packs in a cardboard box).

Pharmacological properties
Pharmacodynamics
Griseofulvin is an antifungal antibiotic. The mechanism of its action is explained by the ability to cause disruption of the structure of the mitotic spindle and cell wall synthesis in chitin-containing fungi, as well as suppress the division of fungal cells in the metaphase and protein synthesis, disrupting the connection with messenger RNA. The substance tends to accumulate in the cells of the skin, nails and hair.
The drug is active against fungi of the genus Microsporum, Trichophyton, Epidermophyton.
Pharmacokinetics
The absorption of griseofulvin is high. The maximum concentration varies in the range from 0.5 to 5 mg / l, is reached within 4-5 hours after ingestion of the drug.
The connection with plasma proteins is 80%. Griseofulvin selectively accumulates in the stratum corneum and its appendages, as well as in skeletal muscles, adipose tissue and liver. A small amount of the substance is distributed in tissues and body fluids. The drug crosses the placenta.
Griseofulvin is metabolized in the liver, resulting in the formation of inactive metabolites (6-methylgriseofulvin and a glucuronide derivative). It is excreted by the kidneys (unchanged - about 1%) and through the intestines (unchanged - about 36%). The half-life is 24 hours.
Indications for use
According to the instructions, Griseofulvin is used in the treatment of infectious diseases:
- Epidermophytosis of hands and feet, caused by Trichophyton interdigitale, Trichophyton rubrum;
- Dermatophytosis of smooth skin, scalp and nails caused by fungi sensitive to the agent, including trichophytosis, microsporia and favus.
Contraindications
- Malignant neoplasms;
- Impaired liver and kidney function;
- Pathology of cerebral circulation, including a history;
- Changes in the morphological composition of the blood;
- Uterine bleeding;
- Porphyria;
- Systemic lupus erythematosus;
- Children under 3 years old;
- The period of pregnancy and breastfeeding;
- Hypersensitivity to griseofulvin.
Instructions for the use of Griseofulvin: method and dosage
Griseofulvin tablets are taken orally, with meals, simultaneously with 1 teaspoon of vegetable oil.
Dosage is prescribed depending on the clinical indications and the patient's age:
- Microsporia: for adults - up to 8 pcs. (1000 mg) per day, for children - at the rate of 21-22 mg per 1 kg of body weight per day. It is recommended to take the tablets before the first negative mycological test every day. Further, the treatment takes place in the same dose according to the scheme: 2 weeks the drug is taken every other day and 2 weeks - with a frequency of 2 times a week;
- Trichophytosis, onychomycosis, favus of the scalp: adult patients weighing up to 50 kg - 5 pcs. (625 mg) per day, with a weight of more than 50 kg - for every 10 kg of weight, add 1 tablet (125 mg). The maximum daily dose is 8 pcs. (1000 mg). For children: in the treatment of trichophytosis and favus - the appointment is carried out at the rate of 18 mg per 1 kg of body weight per day, with onychomycosis - at the rate of 16 mg per 1 kg of the child's weight. Treatment of trichophytosis and favus is carried out similarly to microsporia therapy. With onychomycosis, tablets are taken according to the following scheme: the first month - every day, the second month - every other day, then, until the affected nail is completely replaced with a healthy one - 2 times a week. The general course of treatment should last at least 8 months.
Side effects
The use of Griseofulvin can cause the following side effects:
- From the hematopoietic system: leukopenia, rarely - eosinophilia, leukocytosis;
- From the digestive system: dyspepsia;
- From the nervous system: dizziness, headache, disorientation;
- Allergic reactions: skin rash, urticaria.
Overdose

Throughout the entire period of treatment with Griseofulvin, it is necessary to carry out regular (once every 10-15 days) monitoring of the morphological composition of the blood.
Taking pills should be combined with the use of external antifungal agents.
With dermatophytosis of the scalp, it is necessary to wash your hair with soap and warm water 2 times a week and shave your hair once a week.
With onychomycosis, the dead tissue of the affected nails should be systematically removed using keratolytic agents.
With the development of allergic reactions, it is necessary to prescribe antiallergic therapy, in case of an increase in allergic processes, it is necessary to cancel the drug for 3-4 days. After removing the manifestations of allergy, the drug should be resumed with 1/2 tablet per day, and gradually increasing (within 3-4 days), brought to the therapeutic dose.
When treating children, the simultaneous use of multivitamins is recommended.

Barbiturates and primidone reduce the antifungal effect of Griseofulvin.
Griseofulvin enhances the effect of ethanol. Due to the induction of microsomal liver enzymes, the drug reduces the activity of indirect anticoagulants and estrogen-containing oral contraceptives.
