Procto-Glivenol is a local drug with venotonic, local anesthetic and anti-inflammatory effects, used in the treatment of hemorrhoids.

Release form and composition

Dosage forms of release:

  • rectal cream: white, homogeneous, has a weak characteristic odor (30 g each in aluminum tubes, in a cardboard box 1 tube complete with a polyethylene tip and a latex cap-cap);
  • rectal suppositories: torpedo-shaped, solid, without cracks, somewhat greasy to the touch, yellowish-white, have an unexpressed specific odor (5 pcs. in strips, 2 strips in a cardboard box).

Composition for 100 mg of Procto-Glivenol cream:

  • active substances: tribenoside - 5 mg; lidocaine hydrochloride - 2.12 mg;
  • auxiliary components: stearic acid - 9 mg; sorbitol - 6 mg; propyl parahydroxybenzoate - 0.05 mg; cetyl alcohol - 5 mg; cetomacrogol 1000 - 4.44 mg; isopropyl palmitate - 2 mg; liquid paraffin - 10 mg; methyl parahydroxybenzoate - 0.18 mg; sorbitan stearate - 1.56 mg; water - 54.65 mg.

Composition for 1 suppository Procto-Glivenol:

  • active substances: tribenoside - 400 mg; lidocaine - 40 mg;
  • auxiliary components: witepsol E85 - 305 mg; witepsol W35 - 1255 mg.

Pharmacological properties

Pharmacodynamics

Procto-Glivenol is one of the combined antihemorrhoidal drugs.

The main properties of active ingredients:

  • tribenoside: improves vascular tone, reduces capillary permeability, antagonistically affects some endogenous substances that play the role of mediators in the development of pain and inflammation;
  • lidocaine: has a local anesthetic effect.

The use of Procto-Glivenol leads to a rapid relief of the symptoms of hemorrhoids (pain, itching and skin tension).

Pharmacokinetics

When applying the cream, 2–20% of tribenoside is absorbed. The systemic bioavailability of a substance coming from suppositories reaches 30% of the total amount that is absorbed after ingestion. Cmax (maximum concentration) of tribenoside in blood plasma after rectal administration of 1 suppository is achieved after 2 hours and is 0.001 mg / ml (tribenoside + metabolites).

Tribenoside metabolism occurs mainly in the liver. From 20 to 27% of the dose is eliminated in the form of metabolites in the urine.

Indications for use

According to the instructions, Procto-Glivenol is prescribed for the treatment of external and internal hemorrhoids.

Contraindications

Absolute:

  • liver failure;
  • I trimester of pregnancy;
  • individual intolerance to any component of the drug.

Instructions for the use of Procto-Glivenol: method and dosage

The drug is administered rectally.

Recommended frequency of application:

  • severe clinical symptoms: 2 times a day (morning and evening);

Procto-Glivenol cream is injected using a nozzle (1 tube is usually enough for 20-30 doses). Single dose of suppositories - 1 pc.

Side effects

Possible adverse reactions (> 10% - very common;> 1% and 0.1% and 0.01% and <0.1% - rarely; <0.01% - very rare):

  • immune system: very rarely - anaphylactic reactions, including facial edema, angioedema, cardiovascular disorders and bronchospasm;
  • skin and subcutaneous tissues: rarely - skin reactions at the site of application in the form of burning, rash, urticaria and itching (symptoms may spread beyond the area of application).

Overdose

There is no information on overdose cases.
