Release form and composition
Dosage form - film-coated tablets: cylindrical, convex on both sides, white or white with a cream shade; on the break, a core of white with a gray-cream shade of color is visible (4 and 10 pcs. in blisters, in a cardboard box 1, 2, 3, 4 or 5 packages; 4, 10, 16, 20, 30 and 50 pcs.. in polymeric containers, in a cardboard box 1 container).
Active ingredient: tinidazole, in 1 tablet - 500 mg.
Auxiliary components: potato starch, croscarmellose sodium, microcrystalline cellulose, povidone (polyvinylpyrrolidone), magnesium stearate.
Shell composition: titanium dioxide, macrogol, hypromellose, talc (magnesium hydrosilicate).

Tinidazole is an antiprotozoal agent with antimicrobial action.
The drug is active against Lamblia spp., Trichomonas vaginalis and Entamoeba histolytica. It has a bactericidal effect against anaerobic microorganisms such as Fusobacterium spp., Clostridium spp., Peptostreptococcus spp., Eubacterium spp., Bacteroides spp. (including Bacteroides fragilis and Bacteroides melaninogenicus), Peptococcus spp.
The mechanism of action of tinidazole is explained by its ability to inhibit synthesis and damage the structure of bacterial DNA.
Pharmacokinetics
After oral administration, tinidazole is almost completely absorbed. Its bioavailability is almost 100%. Only 12% binds to plasma proteins.
The maximum concentration in the blood after oral administration of 2 g of tinidazole is 40-51 μg / ml and is reached within 2 hours. After 24 hours, the plasma level of the drug is 11-19 μg / ml, after 72 hours - 1 μg / ml.
The volume of distribution of tinidazole is 50 liters. Penetrates the blood-brain barrier and the placenta. Within 72 hours after administration, it is excreted in breast milk.
It undergoes metabolism in the liver, resulting in the formation of pharmacologically active hydroxylated derivatives that enhance the action of tinidazole and inhibit the growth of anaerobic microorganisms.
The half-life is 12-14 hours. The drug is excreted in the bile - 50%, in the urine - 25% (unchanged) and 12% (in the form of metabolites). Undergoes reabsorption in the renal tubules.
Indications for use
- amoebiasis (including intestinal and hepatic forms);
- giardiasis;
- trichomoniasis (endometritis, colpitis, ovarian and tubo-ovarian abscesses);
- infections caused by anaerobic bacteria (with acute ulcerative gingivitis, lung abscess, pleural empyema, pneumonia; skin and soft tissue infections);
- mixed aerobic-anaerobic infections (in combination with antibiotics);
- eradication of Helicobacter pylori (in combination with antibiotics and preparations containing bismuth).
Tinidazole is also prescribed to prevent postoperative infectious complications caused by anaerobes.
Contraindications
- oppression of bone marrow hematopoiesis;
- organic diseases of the central nervous system;
- children under 12 years old;
- I trimester of pregnancy;
- lactation;
- hypersensitivity to drug components or other 5-nitroimidazole derivatives.
Instructions for the use of Tinidazole: method and dosage
Tinidazole tablets should be taken orally in order to avoid the development of side effects from the digestive system - during or after a meal.
Recommended dosage regimens:
- giardiasis, trichomoniasis: adults - 2000 mg once, children - 50-75 mg / kg once. If necessary, the drug is taken again in the same dose;
- infections in adults caused by anaerobes: the initial dose is 2000 mg once, from day 2 - 1000 mg per day. The course of therapy is 5–6 days;
- intestinal amebiasis: adults - 2000 mg per day for 2-3 days, children - 50-60 mg / kg for 3 days;
- hepatic amoebiasis: adults - 1500–2000 mg per day for 3–6 days, children - 50–60 mg / kg for 5 days.
To prevent the development of postoperative complications in adults, the drug is prescribed 12 hours before the forthcoming surgical intervention in a dose of 2000 mg.
Side effects
- allergic reactions: rash, pruritus, urticaria, angioedema;
- from the central nervous system: ataxia, dizziness, headache, peripheral neuropathy; rarely - convulsions;
- from the gastrointestinal tract: unpleasant taste and dry mouth, diarrhea, nausea, anorexia, vomiting;
- others: rarely - weakness, transient leukopenia.
Overdose
Overdose cases have not been described. There is no specific antidote for the drug. Symptomatic and supportive treatment is recommended. If a little time has passed since taking the drug, gastric lavage may be helpful. Tinidazole is easily removed from the body by dialysis.

During treatment, you should not consume alcoholic beverages, since there is a risk of developing disulfiram-like reactions.
With long-term treatment (more than 6 days), it is necessary to control the peripheral blood picture.
In the case of genital trichomoniasis, both sexual partners should be treated.
Tinidazole makes the urine dark.
Influence on the ability to drive vehicles and complex mechanisms
During the period of therapy, it is recommended to be careful when driving and performing potentially dangerous types of work.
Application during pregnancy and lactation
According to the instructions, Tinidazole is contraindicated in the first trimester of pregnancy. In the II and III trimesters, the use of the drug is possible only in cases where the doctor decides that the expected benefit to the woman outweighs the potential risks to the fetus.
If a therapeutic course is required during lactation, it is recommended to stop breastfeeding, since tinidazole is determined in mother's milk within 72 hours after ingestion.
Pediatric use
Tinidazole tablets are not prescribed for children under 12 years of age, since its safety and efficacy in patients of this age category have not been established.
Drug interactions
Tinidazole is compatible with sulfonamides and antibiotics (rifampicin, erythromycin, aminoglycosides, cephalosporins). Its metabolism is accelerated with the simultaneous use of phenobarbital.
It is not recommended to use ethionamide during treatment.
The drug increases the effect of indirect anticoagulants. To reduce the risk of bleeding, the dose should be halved.
Tinidazole enhances the effect of ethanol.
