Artroker is a non-steroidal anti-inflammatory drug (NSAID), an inhibitor of the activity and synthesis of IL-1 (interleukin-1); inhibits the development of inflammation, degradation and subsequent destruction of cartilage tissue in osteoarthritis.
Release form and composition
Dosage form of the drug - capsules: No. 2, gelatinous hard, with the inscription Micro / Micro, body - gray, lid - blue, filled with yellow granular powder (10 pcs. In aluminum strips or aluminum / aluminum blisters, in a cardboard box 1, 3 or 10 strips or blisters).
1 capsule contains:
- active substance: diacerein - 50 mg;
- auxiliary components: povidone, lactose monohydrate, croscarmellose sodium, magnesium stearate, sodium lauryl sulfate, colloidal silicon dioxide;
- capsule body: gelatin, titanium dioxide, brilliant blue dye, floxin B dye, propyl parahydroxybenzoate, methyl parahydroxybenzoate, sodium lauryl sulfate, water;

Pharmacological properties

Diacerein is an inhibitor of the synthesis and activity of interleukin-1 (IL-1), which plays an important role in the development of inflammation, degradation and subsequent destruction of cartilage in osteoarthritis. Diacerein also helps to slow down the formation of metalloproteinases (collagenases) involved in the processes of damage to cartilage tissue. In the case of prolonged use, the active ingredient stimulates the synthesis of proteoglycans without affecting the synthesis of prostaglandins.
Pharmacokinetics
After oral administration, diacerein is rapidly absorbed from the gastrointestinal tract, completely deacetylated to rhein. In the case of simultaneous administration of the drug with food, bioavailability increases by 25%. The time to reach the maximum concentration of rhein (C max) in blood plasma is 2.4 hours. The half-life is 4.25 hours. The connection between rhein and albumin is about 100%. In the case of a single dose of the drug at a dose of 50 mg, C max is 3.15 mg / l, and with repeated use, C max increases due to the accumulation of the drug. About 20% of rhein is excreted unchanged by the kidneys, 60% in the form of glucuronide, another 20% in the form of sulfate. Rein is able to penetrate the blood-brain and placental barriers.
Indications for use
According to the instructions, Artroker is indicated for use in primary and secondary osteoarthritis.
Contraindications
Absolute:
- severe renal / liver dysfunction;
- inflammatory bowel disease, intestinal obstruction;
- lactase deficiency, lactose intolerance, glucose-galactose malabsorption;
- the period of pregnancy and breastfeeding (lactation);
- children and adolescents up to 18 years old;
- hypersensitivity to anthraquinone glycosides (laxatives of the anthraquinone group - buckthorn bark, senna leaf, etc.);
- increased individual intolerance to the active and / or auxiliary components of the drug.
In special cases, with caution and under medical supervision, NSAIDs are prescribed to patients when diagnosing irritation of the lower intestinal tract.
Instructions for the use of Artroker: method and dosage
Artroker capsules are taken orally after a meal, without chewing (swallowing whole) and drinking water.
Recommended dosage regimen: 1 pc. 2 times a day (after breakfast and after dinner). A significant therapeutic effect is observed after 2-4 weeks from the start of taking the drug.
Artroker should be taken continuously for a long time or in long, at least 4 months, courses. The duration of the break between courses of therapy is determined by the doctor.
Side effects
At the beginning of treatment, negative side effects from the digestive system are possible, such as nausea, vomiting, diarrhea, and abdominal pain. In this case, it is necessary to reduce the dose to 1 capsule per day.
Increased individual sensitivity to the components of the drug can be manifested by hypersensitivity reactions of varying degrees of intensity, rash, itching.
During treatment, depending on the change in pH, an intense coloration of urine from yellow to brown is possible, which does not affect the therapeutic effect and does not require a dose reduction or discontinuation of the drug.

Application during pregnancy and lactation

Drug interactions
