Form of release and composition
Dosage form - capsules: size No. 0, gelatinous solid, with an orange cap and a white body; content - powder from white to light brown or light yellow (20 pcs. in blister strip packaging, in a cardboard box 3 or 6 packs; 60, 90 and 120 pcs. in polymer cans with first opening control, in a cardboard box pack of 1 can).

- active substance: indole carbinol - 200 mg;
- additional components: modified corn starch, lactose monohydrate (milk sugar), magnesium stearate, microcrystalline cellulose.
Pharmacological properties
Pharmacodynamics
The active substance of Indinola Forto is indolecarbinol - a universal corrector of pathological hyperplastic processes in the tissues of the mammary gland. It has antiproliferative and antiestrogenic effects. The main property of the drug is its ability to induce selective death of breast cells with abnormally high proliferative activity.
Indolecarbinol modulates the cytochrome system, resulting in the formation of the cytochrome P450 isoform, CYP1A1, which hydroxylates estrogens in the second position. The resulting metabolite, 2-hydroxyestrone (2-OHE1), is an antagonist of the estrogen receptor and blocks its activation by estrogens themselves and their dangerous metabolites, including 16-α-hydroxyestrone (16α-OHE1). Thus, the induction of estrogen-dependent genes is suppressed, and the cell stops receiving overstimulation.
Indinol Forto also blocks other signaling mechanisms (cytokine), which, by suppressing signaling cascades from the corresponding receptors, stimulate pathological cell growth in breast tissues.
The drug reduces the intensity of pain or relieves pain in the mammary gland with cyclic mastalgia (mastodynia).
Indinol Forto does not contribute to an increase in body weight; it belongs to practically non-toxic drugs (LD 50 > 5 g / kg).
Indications for use
According to the instructions, Indinol Forto is used for cyclic mastalgia (mastodynia), including those developed as a result of benign breast hyperplasia.
Contraindications
- glucose-galactose malabsorption;
- lactase deficiency;
- hereditary galactose intolerance;
- children and adolescents up to 18 years old;
- pregnancy;
- breast-feeding;
- hypersensitivity to drug components.
Instructions for the use of Indinola Forto: method and dosage
Indinol Forto is indicated for oral administration, before meals.
Adult women are prescribed 1 capsule 2 times a day.
The duration of treatment is 6 months.
Side effects
- from the digestive system: pain in the epigastric region;
- from the reproductive system: violation of the menstrual cycle (lengthening or shortening);
- laboratory parameters: eosinophilia, a decrease in the concentration of creatinine, an increase in the concentration of estradiol, prolactin, follicle-stimulating and thyroid-stimulating hormones;
- other: weight loss.
According to the data of clinical studies, the incidence of side effects in patients receiving Indinol Forto was approximately comparable to that in women receiving placebo (26.9% and 20%, respectively). The undesirable reactions that arose were of a mild nature, passed on their own or were stopped by the appointment of symptomatic agents.
Overdose
In case of overdose, diarrhea, nausea and vomiting may occur.
The specific antidote for indolecarbinol is unknown. Treatment is symptomatic.
