Genferon is a local and systemic immunomodulatory drug with antioxidant, immunomodulatory, local anesthetic and antiviral effects.

Release form and composition

Genferon is produced in the form of suppositories: a cylindrical shape with a pointed end, a homogeneous structure, from white to white with a yellow tint, the presence of a funnel-shaped depression or a hollow rod on the cut is possible (5 pcs. In a blister strip, in a cardboard box 1 or 2 packages).

Content of active ingredients in 1 suppository:

  • Recombinant human interferon alpha-2 - 250 thousand, 500 thousand or 1 million IU;
  • Taurine - 10 mg;
  • Benzocaine - 55 mg

Excipients: dextran 60,000, tween-80, sodium citrate, polyethylene oxide 1500, citric acid, T2 emulsifier, solid fat, purified water.

Pharmacological properties

Pharmacodynamics

The active component of Genferon is recombinant human interferon alpha-2b, synthesized by a strain of the bacterium Escherichia coli, modified using genetic engineering methods, allowing to introduce the human interferon alpha-2b gene into it.

Interferon alfa-2b is characterized by antiviral, antibacterial, antiproliferative and immunomodulatory effects. The antiviral effect is associated with the activation of some intracellular enzymes that inhibit viral replication. The immunomodulatory effect is expressed primarily in the stimulation of cell-mediated reactions of the immune system, which enhances the effectiveness of the immune response against cells undergoing tumor transformation, intracellular parasites and viruses. This is possible due to the activation of phagocytosis and the monocyte-macrophage system, increased differentiation of B-lymphocytes and their synthesis of antibodies, stimulation of the activity of NK-cells (natural killers) and CD8 + T-killers, as well as an increase in the expression of molecules that make up the main histocompatibility complex of type I,which increases the chances of recognition of infected cells by cells of the immune system. Since interferon enhances the functions of leukocytes localized in all layers of the mucous membrane, this allows them to actively participate in the elimination of pathological foci. Also, under the influence of interferon, the production of secretory immunoglobulin A is normalized. The antibacterial effect is due to the reactions of the immune system, which are enhanced by interferon.which is enhanced by interferon.which is enhanced by interferon.

Taurine normalizes metabolic processes and ensures tissue regeneration, and is also characterized by an immunomodulatory and membrane-stabilizing effect. This active component of Genferon prevents the occurrence of pain impulses in the endings of the nerves with sensitivity and their transmission along the nerve fibers. Benzocaine has an exclusively local effect and does not enter the systemic circulation.

Pharmacokinetics

With rectal administration of Genferon, there is a high bioavailability of interferon, exceeding 80%, which explains both local and pronounced systemic immunomodulatory effects. With intravaginal use, due to the high concentration of this component in the focus of infection and its fixation on the cells of the mucous membrane, a pronounced local antibacterial, antiproliferative and antiviral effect is observed. Since the vaginal mucosa has a low absorption capacity, the systemic effect of interferon is insignificant.

The maximum level of interferon in the blood serum is recorded 5 hours after the administration of Genferon. The elimination of this substance from the body is mainly carried out by renal catabolism. The half-life is 12 hours, which is due to the frequency of application of the drug 2 times a day.

Indications for use

According to the instructions, Genferon is indicated for use as part of the complex treatment of such diseases / conditions:

  • Acute bronchitis;
  • Infectious and inflammatory diseases of the urogenital tract: chlamydia, genital herpes, ureaplasmosis, recurrent vaginal candidiasis, mycoplasmosis, gardnerellosis, human papillomavirus infection, trichomoniasis, bacterial vaginosis, erosion of the cervix, vulvovaginitis, ballexolethycinitis, ureteritis, ureteritis, balvovaginitis, ballexolrethynostanitis,

Contraindications

Hypersensitivity to the components of the drug is the only contraindication to its use.

Instructions for the use of Genferon: method and dosage

Genferon is used intravaginally and / or rectally:

  • Infectious and inflammatory diseases of the urogenital tract of men: rectally, 1 suppository (depending on the severity of the condition, 500,000 or 1,000,000 IU), 2 times a day for 10 days;
  • Infectious and inflammatory diseases of the urogenital tract of women: rectally or intravaginally (depending on the nature of the disease), 1 suppository 250,000, 500,000 or 1,000,000 IU (depending on the severity of the condition) 2 times a day, for 10 days daily. In the case of protracted illnesses, 1 suppository 3 times a week (every other day), duration - 1-3 months. For the treatment of a pronounced infectious-inflammatory process in the vagina - 1 suppository 500,000 IU intravaginally in the morning and 1 suppository 1,000,000 IU rectally at night, at the same time a suppository with antibacterial / fungicidal agents should be inserted into the vagina;
  • Normalization of local immunity indices in women at 13-40 weeks of gestation during the treatment of infectious and inflammatory diseases of the urogenital tract: 1 suppository 250,000 IU intravaginally 2 times a day, for 10 days daily;
  • Chronic recurrent cystitis (as part of complex therapy) in adults: in case of exacerbation - in combination with a standard course of antibiotic therapy, 1 suppository 1,000,000 IU rectally 2 times a day for 10 days, then every other day, for 40 days, in that the same dose for the prevention of relapse;
  • Acute bronchitis (as part of complex therapy) in adults: 1 suppository 1,000,000 IU rectally 2 times a day for 5 days.

Side effects

The use of the drug can cause the development of allergic reactions in the form of itching, skin rash, burning sensation in the vagina. Side effects are reversible and disappear within 72 hours after discontinuation or dose reduction.

The temperature may rise after drug administration.

With a daily dose of 10 million IU, the following side effects are highly likely:

  • From the hematopoietic system: thrombocytopenia, leukopenia;
  • From the central nervous system: headache;
  • Others: increased sweating, fever, loss of appetite, weakness, myalgia, arthralgia.

Overdose

Cases of Genferon overdose in clinical practice are almost unknown. In case of accidental single administration of suppositories in a larger amount than was prescribed by the doctor, it is necessary to suspend further administration of the drug for at least 24 hours, after which you can return to the original treatment regimen.
