Release form and composition

The drug is produced in the form of film-coated tablets: white, round, biconvex, engraved on one side "ANA 1", a white or almost white core (in blisters of 10 pcs., In a cardboard box of 3 or 9 blisters; in blisters of 14 pcs., in a cardboard box 2 blisters. Each box also contains instructions for the use of Anzales).

1 tablet contains:

  • active substance: anastrozole - 1 mg;
  • auxiliary components: lactose monohydrate, sodium carboxymethyl starch, povidone, magnesium stearate;
  • film shell: White opadrai (hypromellose, macrogol 400, titanium dioxide).

Pharmacological properties

Pharmacodynamics

Anzales is an antineoplastic drug. Its active ingredient - anastrozole, being a selective non-steroidal aromatase inhibitor, helps to reduce the concentration of circulating estradiol and has a therapeutic effect in women with breast cancer. The mechanism of action of the drug is due to the fact that postmenopausal estradiol is predominantly formed from estrone produced in peripheral tissues by conversion with the participation of the aromatase enzyme from androstenedione. Taking anastrozole in postmenopausal women at a dose of 1 mg per day reduces estradiol levels by 80%.

The drug does not possess estrogenic, progestogenic and androgenic activity. Therapeutic doses of anastrozole do not affect the secretion of cortisol and aldosterone.

Pharmacokinetics

After oral administration, anastrozole is rapidly absorbed from the gastrointestinal tract. When used on an empty stomach, the maximum plasma concentration (C max) is reached within 2 hours. The presence of food in the stomach slightly reduces the rate, but does not affect the degree of absorption.

Plasma protein binding - 40%.

Metabolized by N-dealkylation, hydroxylation and glucuronidation. The main metabolite of anastrozole is triazole, it does not inhibit aromatase.

It is excreted mainly through the kidneys within 72 hours after taking a single dose, including in unchanged form - less than 10%. The half-life (T 1/2) from plasma is 40-50 hours. There is no information on cumulation.

With stabilized liver cirrhosis or impaired renal function, the clearance of anastrozole does not change.

Indications for use

The use of the drug Anzales is indicated for the treatment of postmenopausal breast cancer.

Contraindications

Absolute:

  • premenopause;
  • severe liver failure;
  • concomitant use of tamoxifen or drugs containing estrogens;
  • period of pregnancy and lactation;
  • childhood;
  • hypersensitivity to the components of the drug.

Anzales should be taken with caution, preferably under medical supervision, with osteoporosis, hypercholesterolemia, ischemic heart disease, mild to moderate liver dysfunction, severe renal failure (creatinine clearance less than 20 ml / min), lactase deficiency, lactose intolerance, glucose-galactose malabsorption (this dosage form contains lactose).

Anzales tablets are taken orally, swallowing whole and drinking plenty of water, regardless of the meal, but always at the same time of day, convenient for the patient.

Recommended dosage for adults, including elderly patients: 1 pc. (1 mg) once a day, the drug is taken for a long time.

If signs of progression of the disease appear, taking the tablets should be discontinued. The optimal duration of adjuvant therapy is 5 years.

Side effects

  • from the central nervous system: drowsiness, headache, asthenia;
  • from the digestive system: nausea, vomiting, diarrhea, anorexia; with advanced breast cancer (mainly with liver metastases) - an increase in the level of alkaline phosphatase (alkaline phosphatase) and more often - GGT (gamma-glutamyltransferase);
  • from the endocrine system: flushing of the face, hair thinning, vaginal dryness;
  • from the side of metabolism: a slight increase in the level of total cholesterol;
  • allergic reactions: skin rash.
