Pharmacological properties

Pharmacodynamics

Mebeverin is a myotropic antispasmodic drug. Its active ingredient is mebeverine hydrochloride, the direct action of which is directed to the smooth muscles of the gastrointestinal tract. The ability of the drug to block the passage of sodium ions into smooth muscle cells allows you to eliminate spasm without affecting normal intestinal motility. It does not cause reflex intestinal hypotension, does not have anticholinergic activity.

Pharmacokinetics

After oral administration, mebeverine is rapidly absorbed, the degree of absorption is high. The relative bioavailability is 97%. The prolonged form of the drug provides a clinical effect when using the dosage regimen 2 times a day.

Against the background of taking repeated doses, a significant cumulation of the drug does not occur.

Mebeverine is completely metabolized in the human body. Its metabolism mainly occurs with the help of esterases, which in the first stage break down the ether into mebeverine alcohol and veratric acid. The main metabolite of mebeverine circulating in blood plasma is demethylated carboxylic acid. When taking the drug 200 mg 2 times a day, the maximum concentration (C max) of demethylated carboxylic acid in the blood plasma is reached within about three hours and is 804 ng / ml. The half-life in the equilibrium state of the main metabolite is approximately 5.77 hours.

The drug is excreted from the body almost completely in the form of metabolites through the kidneys.

Elderly patients, as well as patients with impaired renal and / or liver function, do not require dose adjustment.
