Release form and composition
- Capsules: gelatinous, yellow-beige, matte, with the inscription "Venoruton 300", odorless; content - odorless powder, yellow or yellowish-brown color (10 pcs. in blisters, in a cardboard box 2 or 5 blisters);
- Gel for external use 2%: homogeneous, transparent, golden yellow color, sometimes slightly opalescent, practically odorless (40 and 100 g each in aluminum tubes, 1 tube in a cardboard box).
Active ingredient: hydroxyethylrutosides, in 1 capsule - 300 mg, in 1 g of gel - 20 mg.
Additional components of capsules:
- Excipients: macrogol 6000, titanium dioxide, iron dye yellow dioxide, gelatin;
- Ink composition: propylene glycol, shellac, water, n-butanol, isopropanol, iron dye black dioxide.
Excipients of the gel: disodium EDTA, sodium hydroxide 30%, benzalkonium chloride, carbomer, purified water.
Pharmacodynamics and pharmacokinetics
The drug is a rutin derivative. It affects the capillaries and veins, strengthening their walls and increasing tone, and generally improves blood circulation.
Acting on the fibrous matrix located between the endothelial cells, Venoruton narrows the intercellular pores.
By inhibiting the aggregation of erythrocytes and increasing the degree of their deformability, the drug causes an anti-inflammatory effect.
Venoruton reduces the severity of pain, edema, trophic disorders, seizures and varicose ulcers in chronic venous insufficiency; relieves the symptoms of hemorrhoids (itching, pain, bleeding).
The beneficial effect of the drug on the resistance and permeability of the capillary walls slows down the development of diabetic retinopathy, and due to the effect on the rheological properties of the blood, retinal microthrombosis is prevented.
The maximum plasma concentration of the drug is reached within 2-9 hours. The concentration remains measurable for 40 hours. 27-29% of the drug binds to plasma proteins. The active substances and their metabolites are excreted mainly in the bile, about 3–6% are excreted by the kidneys within 48 hours. The half-life is 13.5-25.7 hours, but this value is constant in each patient.
When applied topically, the active ingredients quickly penetrate the epidermal barrier: after 30 minutes they appear in the dermis, and after 2-5 hours - in the layer of subcutaneous adipose tissue. When using Venoruton gel, the concentration of active ingredients in the blood plasma is low and is not determined by the currently available methods for measuring the plasma concentration of substances.
Indications for use
For capsules:
- Trophic disorders in varicose veins;
- Postphlebitic syndrome;
- Chronic venous insufficiency;
- Trophic ulcers;
- Hemorrhoids (exudation, itching, pain and bleeding);
- Lymphostasis after removal of varicose veins and sclerotherapy (as an auxiliary drug);
- Retinopathy in patients with diabetes mellitus, arterial hypertension and atherosclerosis (as adjunctive therapy);
- Hemorrhoids and venous insufficiency in pregnant women (starting from the second trimester).
For the gel:
- Pain and heaviness in the legs, swelling of the ankles;
- Swelling and pain associated with chronic venous insufficiency;
- Edema and pain of traumatic origin (including with bruises and sprains of muscles, damage to ligaments);
- Pain after sclerotherapy.
Contraindications
- I trimester of pregnancy;
- Hypersensitivity to drug components.
Instructions for the use of Venoruton: method and dosage
The capsules should be taken orally with meals with plenty of water. The initial dose is 300 mg 3 times a day. After 2 weeks of treatment, the drug is canceled or the dose is reduced to the minimum maintenance dose - 600 mg per day, if necessary, the dose is left unchanged.
With diabetic retinopathy, the daily dose is 900-1800 mg, with lymphostasis - 3000 mg.
According to the instructions, Venoruton gel is applied to the affected area, rubbing gently until completely absorbed, 2 times a day - in the morning and in the evening. If necessary, the drug can be applied under elastic stockings or bandages.
Side effects
When using Venoruton in capsule form, the following side effects are possible (usually disappear after the drug is discontinued):
- Digestive system: heartburn, nausea, and diarrhea;
- Allergic reactions: skin rash;
- Others: flushing of the face, headache.
When using Venoruton in the form of a gel, local skin reactions are possible due to increased sensitivity to the components of the drug.
Overdose
No cases of overdose accompanied by clinical symptoms were recorded.

Application during pregnancy and lactation
In pregnant women, the use of the drug was studied in clinical trials only in the II and III trimesters. In the first trimester, the use of the drug is contraindicated.
Studies carried out on laboratory animals have not revealed teratogenic and other harmful effects on the fetus.
Venoruton in the form of capsules can be used no earlier than the second trimester of pregnancy and only in cases where the benefit to the mother outweighs the potential risk to the fetus.
Drug interactions
The action of Venoruton, taken orally, is enhanced by ascorbic acid.
There is no information on the interaction of the drug with other drugs when used externally.
