Axamon is a cholinesterase inhibitor, an agent for the treatment of disorders of the nervous system.

Release form and composition

Axamon is produced in tablets and in solution for subcutaneous or intramuscular administration.

The active ingredient in all forms of the drug is ipidacrine hydrochloride in the form of a monohydrate. 1 tablet contains 15 mg of active ingredient. Excipients in the tablets are sodium carboxymethyl starch, ludipress and calcium stearate. The solution contains 0.1M hydrochloric acid solution and water for injection.

Pharmacological properties

Pharmacodynamics

Axamon is a reversible cholinesterase inhibitor that blocks membrane potassium channels, directly stimulating impulse conduction in the central nervous system and neuromuscular synapse. Enhances the action of acetylcholine, adrenaline, histamine, oxytocin and serotonin on smooth muscles.

Axamon has the following pharmacological effects:

  • improvement and stimulation of impulse conduction in the central nervous system and neuromuscular transmission;
  • improvement of conductivity in the peripheral nervous system in case of disorders due to inflammation, trauma, exposure to local anesthetics, potassium chloride, some antibiotics, etc.;
  • increased contractility of smooth muscle organs under the influence of agonists of adrenaline, acetylcholine, histamine, oxytocin and serotonin receptors (excluding potassium chloride);
  • improvement of memory, inhibition of the progressive development of dementia.

Axamon has no embryotoxic, teratogenic, carcinogenic, immunotoxic and mutagenic effects. Does not affect the endocrine system.

Pharmacokinetics

When taken orally, it is rapidly absorbed from the gastrointestinal tract. The time to reach the maximum plasma concentration is 60 minutes. The connection with plasma proteins is 40–55%. The half-life is about 40 minutes. The active substance is metabolized in the liver. It is excreted through the kidneys (the main route is tubular secretion, 1/3 of the dose is excreted by glomerular filtration) and extrarenally.

Indications for use

According to the instructions, Axamon is prescribed for the treatment of:

  • Polyneuritis;
  • Neuritis;
  • Myasthenia gravis and myasthenic syndromes;
  • Multiple sclerosis;
  • Myelopolyradiculoneuritis;
  • Neuropathy;
  • Paresis and bulbar paralysis, accompanied by movement disorders;

With myasthenia gravis, myasthenic syndrome, demyelinating lesions of the nervous system, an increased dosage is used: 20-40 mg 4-5 times a day. The maximum allowable daily dosage is 180-200 mg.

For the treatment of Alzheimer's disease and other types of dementia, the dosage is selected individually, on average it is 10-20 mg 2-3 times a day. This takes into account the fact that no more than 200 mg of the drug can be administered per day. The duration of treatment is from 1 to 12 months.

When prescribing the drug in high doses, it is recommended to take it 4-5 times a day with a break of 2-3 hours.

With intestinal atony, 15-20 mg of the drug is prescribed 2-3 times a day for 1-2 weeks.

Side effects

The drug is well tolerated in most cases. In some cases, the use of Axamon causes:

If β-blockers were used before the start of Axamon therapy, the risk of developing bradycardia increases.

Weakens the inhibitory effect of local anesthetics, potassium chloride, aminoglycosides on the conduction of excitation along peripheral nerves and neuromuscular transmission.

Drinking alcohol increases the side effects of Axamon.
