Spazoverin is a myotropic antispasmodic agent.

Release form and composition

  • tablets: biconvex, round, yellow (10 pcs. in blisters, in a cardboard box 1, 2, 3, 4, 5 or 10 blisters; 30, 50 or 100 pcs. in polystyrene containers with a polyethylene lid, in a cardboard box 1 container);
  • solution for injection (1 or 2 ml in dark glass ampoules, in a cardboard box of 5 or 25 ampoules).

Each pack also contains instructions for the use of Spazoverin.

Composition of 1 tablet:

  • active substance: drotaverine hydrochloride - 40 or 80 mg;
  • auxiliary components: microcrystalline cellulose, starch, magnesium stearate, polyvinylpyrrolidone, talc.

Composition of 1 ml solution for injection:

  • active substance: drotaverine hydrochloride - 20 mg;
  • auxiliary components: disodium edetate, sodium metabisulfite, water for injection, ethyl alcohol.

Pharmacological properties

Pharmacodynamics

The active substance of Spazoverin - drotaverine hydrochloride, is a myotropic antispasmodic, which is close to papaverine in chemical structure and pharmacological properties, but has a more pronounced and lasting effect.

Drotaverine reduces the flow of calcium into smooth muscle cells (inhibits phosphodiesterase, leads to the accumulation of intracellular cyclic adenosine monophosphate). Reduces intestinal motility and smooth muscle tone of internal organs, dilates blood vessels. Does not penetrate the central nervous system and does not interact with the autonomic nervous system.

Since drotaverine has a direct effect on smooth muscles, it can be used as an antispasmodic in cases where the use of drugs from the group of m-anticholinergic antagonists is contraindicated (for example, in angle-closure glaucoma or prostate adenoma).

Pharmacokinetics

After oral administration, drotaverine is well absorbed, the half-absorption period is 12 minutes.

The antispasmodic is 100% bioavailable. The maximum concentration in the blood reaches after 2 hours. It is evenly distributed over the tissues, penetrates into smooth muscle cells. It binds to plasma proteins by 95–98%. Does not penetrate the blood-brain barrier.

The substance is excreted mainly with the kidneys, to a lesser extent - with bile.

Indications for use

  • functional states and pain syndrome associated with smooth muscle spasm in diseases such as gastric ulcer and duodenal ulcer, spastic colitis, chronic cholecystitis, chronic gastroduodenitis, cholelithiasis, postcholecystectomy syndrome, cardio- and pylorospasm (for prevention and treatment);
  • kidney stone disease;
  • spasms of peripheral arterial and cerebral vessels;
  • algodismenorrhea;
  • labor (to relieve cervical spasm and weaken uterine contractions during childbirth);
  • carrying out instrumental methods for diagnosing organs and systems.

Contraindications

Absolute:

  • arterial hypotension;
  • cardiogenic shock;
  • atrioventricular block II – III degree;
  • severe renal, hepatic, heart failure;
  • hypersensitivity to any component of the Spazoverin solution or tablets.

Relative:

  • severe atherosclerosis of the coronary arteries;
  • glaucoma;
  • prostate adenoma;
  • the period of pregnancy and lactation.

Spazoverin, instructions for use: method and dosage

Spazoverin tablets should be taken orally.

In the form of a solution for adult patients, Spazoverin is administered subcutaneously or intramuscularly at 40–80 mg (2–4 ml) 1 to 3 times a day. The duration of treatment is 1-2 weeks, if necessary, the course is extended. If a longer treatment is required, then they switch to taking the drug inside.

To accelerate the achievement of the effect, you can slowly intravenously inject 2-4 ml of the drug in 10-20 ml of isotonic sodium chloride 0.9% solution or 5% dextrose solution.

Recommended dosage for all forms of Spazoverin release:

  • children under 6 years old - 10–20 mg 1-2 times a day;
  • children from 6 to 12 years old - 20 mg 1-2 times a day;
  • adolescents over 12 years old and adults - 40-80 mg 2-3 times a day.

Side effects

In some cases, the drug causes the following side effects: sweating, fever, palpitations, arterial hypotension, dizziness, allergic skin reactions.

Overdose

When used in high doses, Spazoverin reduces the excitability of the heart muscle and disrupts atrioventricular conduction, can cause paralysis of the respiratory center and cardiac arrest.

  • enhances hypotension caused by procainamide, quinidine, tricyclic antidepressants.

Phenobarbital increases the severity of the antispasmodic effect of drotaverine.
