Ladisan is an antidepressant.

The composition and form of release of Ladisan

Ladisan is produced in two dosage forms:

• Round coated tablets containing 25 or 50 mg of the active ingredient - maprotiline hydrochloride. Additional components of the tablets are: corn starch, lactose, calcium phosphate, talc, gelatin, colloidal silicon dioxide, dyes, hypromellose, polysorbate-80. In blisters of 10 tablets.
• Colorless transparent solution for infusion and intravenous injection. 1 ml of solution contains 5 mg of maprotiline methanesulfonate. The auxiliary components of the solution are: water for injection, methanesulfonic acid and mannitol. In ampoules of colorless glass, 5 ml.

Pharmacological action Ladisan

According to the instructions, Ladisan is a drug belonging to the group of serotonin and norepinephrine reuptake inhibitors. The pharmacological action of maprotiline (a tetracyclic antidepressant) is to stop the reverse neuronal uptake of the neurotransmitters serotonin and norepinephrine by the presynoptic membranes of neurons in the cerebral cortex.

This medicine has an optimal combination of all the main properties of antidepressants. The use of Ladisan raises the mood, promotes the disappearance of an excited state and feelings of fear, relieves psychomotor retardation and various somatic disorders characteristic of masked depression.

Ladisan acts quite quickly, and the therapeutic effect can be observed already several days after the start of treatment. In addition, according to reviews, Ladisan is very well tolerated by the body, therefore it is widely used in outpatient practice.

Indications for Ladisan's use

According to the instructions, Ladisan should be used for various depressions:

• somatogenic (symptomatic, organic);
• endogenous (involutional, cyclic, periodic);
• depression of the climacteric period;
• psychogenic (neurotic, reactive, depressions of exhaustion);
• masked forms of depression.

Ladisan is also prescribed for other depressive disorders, accompanied by the following symptoms:

• apathy;
• irritability or dysphoria;
• feeling of fear.

According to reviews, Ladisan has a beneficial effect on somatic and psychosomatic disorders that arise against the background of a feeling of fear or longing.

Cautions

The drug is prescribed with caution to patients with impaired liver or kidney function, with increased intraocular pressure, with urinary disorders (for example, in diseases of the prostate gland).

With long-term use of Ladisan, patients should regularly conduct an ECG and monitor heart function. This is especially true for patients who suffer from cardiovascular diseases, and elderly patients. Patients prone to orthostatic hypotension should have their blood pressure checked regularly while taking the medication.

It should be borne in mind that, like other psychotropic drugs, the use of Ladisan reduces alcohol tolerance.

According to reviews, Ladisan in tablet form can adversely affect psychophysical reactions, especially if the drug is combined with alcohol or drugs that have a depressive effect on the central nervous system.

Method of application and dosage regimen of Ladisan

The dosage of the drug is selected individually for each patient, depending on the severity of the disease. An increase or decrease in the evening dosage is allowed by decreasing or increasing the daily dose. It is also possible to take the daily dose in one dose, preferably in the evening. When a positive effect is achieved, you can try to reduce the dosage of the drug, but if repeated signs of depression appear, it is recommended to return to the previous dose.

Ladisan tablets are taken inside. Mostly they are used in the outpatient treatment of depression, 25 mg up to three times a day, or 25-75 mg once in the evening. If depression is severe and its treatment is carried out in a hospital setting, then an increase in the daily dose to 150 mg is allowed.

The solution is mainly prescribed for newly diagnosed cases of severe depression or if treatment with Ladisan tablets did not bring the desired result. The daily dosage for intravenous drip infusion is 25-100 mg. To prepare the solution, dissolve the contents of 1-2 ampoules in 250 ml of glucose or sodium chloride solution and inject slowly over two hours. If the patient needs to enter a higher dose of the drug, the infusion time is increased to three hours.

As a rule, after the first two weeks of treatment with Ladisan with the help of infusion therapy, the patient is transferred to oral treatment.

Contraindications

According to the instructions, Ladisan is contraindicated in the following cases:

• hypersensitivity to the main active ingredient - maprotiline;
• acute myocardial infarction;
• convulsions;
• first trimester of pregnancy;
• arrhythmias of the heart.

Use of Ladisan during pregnancy and breastfeeding

In the first trimester of pregnancy, the use of the drug is strictly prohibited. In general, during pregnancy, taking Ladisan is not recommended, except for exceptional indications or in the absence of an alternative.

Since the active ingredient of the drug penetrates into breast milk, breastfeeding should be discontinued during the use of the drug.

Side effects

According to reviews, Ladisan in the first days of its use can cause fatigue and anticholinergic effects (constipation, dry mouth, dizziness, impaired accommodation). In rare cases, tachycardia and low blood pressure are observed. Sometimes skin reactions associated with hypersensitivity to maprotylene and requiring discontinuation of the drug are possible.

Overdose

According to the instructions, Ladisan may cause an overdose. Symptoms develop about four hours after taking the medication and reach a maximum one day later.

Overdose symptoms include:

• disorders of the cardiovascular system (arrhythmia, tachycardia, heart failure, arterial hypotension);
• excitement or depression of the central nervous system (anxiety, convulsions, drowsiness, muscle rigidity, coma, stupor).

In rare cases, an overdose can provoke vomiting, cyanosis, fever, anuria, oliguria, mydriasis, increased sweating.

In case of drug overdose, symptomatic treatment is necessary.

Storage conditions Ladisan

Ladisan in the form of a solution and tablets should be stored at room temperature in a dry place out of the reach of children. The shelf life is 5 years.
