Release form and composition
The drug is produced in the form of film-coated tablets, biconvex, round; film shell and core in a cross section of white or almost white color [in blisters made of printed varnished aluminum foil and polyvinyl chloride (PVC) film, 10, 15 or 20 tablets, in a cardboard box together with instructions for use of Gastrostat 1-6 or 10 packages].
Composition of one film-coated tablet:
- active ingredient: rebamipide - 100 mg;
- additional ingredients: hydroxypropyl methylcellulose (hypromellose), sodium lauryl sulfate, magnesium stearate, magnesium carbonate, collidone CL-F, citric acid;
- film shell: polyethylene glycol 6000 (macrogol 6000), hydroxypropyl methylcellulose (hypromellose), talc, titanium dioxide.
Pharmacological properties
Pharmacodynamics
The active substance in Gastrostat is a gastroprotective agent, the intake of which increases the level of PGE2 (prostaglandin E2) in the gastric mucosa and the level of PGE2 and PGI2 in the contents of gastric juice.
It has a cytoprotective effect on the gastric mucosa when it is damaged by ethanol, ASA (acetylsalicylic acid), alkalis and acids.
When Gastrostat is taken, the mucus content on the surface of the stomach wall increases and enzymes are activated, which accelerate the biosynthesis of high molecular weight glycoproteins.
Rebamipide therapy has a positive effect on the dynamics of the disease, since it occurs:
- increased proliferation and exchange of gastric epithelial cells;
- improving the blood supply to the gastric mucosa;
- cleansing the mucous membrane from hydroxyl radicals;
- activation of alkaline gastric secretion and mucosal barrier function;
- suppression of superoxides produced by polymorphonuclear leukocytes and neutrophils in the presence of Helicobacter pylori;
- protection of the gastric mucosa from damage by bacteria;
- providing a gastroprotective effect when exposed to NSAIDs (non-steroidal anti-inflammatory drugs) on the gastric mucosa.
Pharmacokinetics
Rebamipide, when taken orally, demonstrates a high degree of absorption.
The peak concentration (C max) of the substance in the blood plasma after taking at a dose of 100 mg is observed after ~ 2 hours, the C max is 340 ng / ml.
In experiments in vitro it was found that the drug's connection with plasma proteins varies within the range of 98.4–98.6%.
The half-life is about 1 hour. With repeated doses of Gastrostat, its cumulation in the body is not observed. Rebamipide (~ 10% of the dose taken) is excreted in the urine, mainly unchanged. When the drug is taken at a dose of 600 mg, traces of the hydroxylated metabolite can be isolated.
Indications for use
Gastrostat is used in the following cases:
- treatment of chronic gastritis with increased acid-forming function of the stomach in the acute stage;
- treatment of erosive gastritis;
- therapy of gastric ulcer;
- prevention of the development of mucosal damage while taking NSAIDs.
Contraindications
Absolute:
- children and adolescents under 18 years of age;
- gestation period;
- breastfeeding (lactation);
- individual intolerance to rebamipide or auxiliary components that make up Gastrostat.
The drug is prescribed with caution to elderly patients because of the possible development of hypersensitivity to rebamipide.
Gastrostat, instructions for use: method and dosage
Gastrostat tablets are taken orally with a little water or any other liquid.
Recommended dosage: 1 pc. 3 times a day. The course of admission is from 14 to 28 days, if it is necessary to prolong therapy, its duration can be increased to 56 days.
There are no peculiarities of the drug action when it is first taken or canceled.
If for any reason one dose was missed, the next dose must be taken at the scheduled time. It is not recommended to double the dose in such a situation.
Side effects
- hematopoietic system: granulocytopenia, leukopenia;
- digestive tract: abdominal pain, constipation / diarrhea, flatulence, heartburn, nausea / vomiting, impaired taste;
- liver and biliary tract: increased ACT (serum glutamic aspartate aminotransferase) and ALT (serum glutamic alanine aminotransferase), signs of liver dysfunction;
- allergic reactions: eczematous rash, skin rash, itching;
- others: algodismenorrhea (menstrual irregularities).
Overdose
There is no information about the symptoms of an overdose of rebamipide, since until now there have been no reports of cases of deliberate administration of the drug in ultrahigh doses. If you accidentally take an excessive dose of rebamipide, you may experience nausea, vomiting, constipation or diarrhea, abdominal pain, and headache.
There is no specific antidote. A patient with signs of intoxication should have a gastric lavage and symptomatic therapy.

Influence on the ability to drive vehicles and complex mechanisms
The influence of Gastrostat on the speed of psychomotor reactions has not been studied, therefore, if it is necessary to engage in potentially hazardous activities that require increased concentration of attention, including driving and working with complex mechanisms, caution should be exercised.
Application during pregnancy and lactation
Gastrostat is not prescribed for pregnant women, since the safety of its use in this category of patients has not been proven.
The ability of rebamipide to penetrate a woman's breast milk is the reason for stopping breastfeeding if drug therapy is needed.
Pediatric use
Gastrostat tablets are not prescribed for patients under 18 years of age.
With impaired renal function
There are no special instructions on the dosage regimen for patients with impaired renal function.
For violations of liver function
There are no special instructions on the dosage regimen for patients with impaired hepatic function.
Use in the elderly
Elderly patients should be careful when using Gastrostat for the first time due to the possibility of developing hypersensitivity to the active ingredient of the drug, rebamipide.
