Pharmacological properties

Pharmacodynamics

Pancreoflat is a combined enzyme agent that compensates for the insufficiency of the exocrine function of the pancreas and reduces flatulence. Contains pancreatin and dimethicone as active substances.

Pancreatin is a porcine pancreatic powder containing various enzymes including lipase, alpha-amylase and trypsin.

Lipase cleaves fatty acids at positions 1 and 3 of triglyceride molecules. With this cleavage, free fatty acids are formed, which are absorbed from the upper part of the small intestine mainly with the participation of bile acids.

Alpha-amylase breaks down polysaccharides containing glucose.

Trypsin is formed from trypsinogen in the small intestine by enterokinase. This enzyme cleaves bonds between peptides, in which arginine or lysine was mainly involved. In clinical studies, it has been found that trypsin suppresses pancreatic secretion through a feedback mechanism. It is believed that the analgesic effects of pancreatin, described in some studies, are associated with this.

Dimethicone - the second active component of Pancreoflat - eliminates the increased accumulation of gases in the small intestine. This substance is chemically inert, its mechanism of action is based on the ability to change the surface tension of gas bubbles in the intestine. As a result, the bubbles burst, and the gas in them is released and then sucked in or excreted in a natural way.

Pharmacokinetics

Pancreatin and dimethicone are not absorbed from the gastrointestinal tract.

Indications for use

  • Chronic pancreatitis, gastric achilles and other diseases against the background of insufficient exocrine pancreatic function;
  • Digestive disorders associated with diseases of the liver and biliary tract;
  • Disorder of the digestive process after surgery on the stomach and small intestine, especially with flatulence and other pathologies with increased formation of gases and their accumulation in the intestines.

Contraindications

  • Age under 12;
  • Individual intolerance to the components of the drug.

According to the instructions, Pancreoflat should be prescribed with caution to patients with an early stage of acute pancreatitis, exacerbation of the chronic form of pancreatitis, galactose intolerance, lactase deficiency and impaired absorption of glucose-galactose, during pregnancy and breastfeeding.

Instructions for the use of Pancreoflat: method and dosage

Pancreoflat tablets are taken orally, swallowed whole and washed down with water, during or after meals.

The dose and duration of treatment is determined by the doctor based on clinical indications.

The dosage regimen for adult patients is usually 1-2 tablets with each meal.

Side effects

  • Digestive system: nausea, vomiting, abdominal pain, diarrhea, stomach discomfort, constipation, hyperuricosuria, an increase in uric acid in the blood;
  • Dermatological reactions: in case of hypersensitivity, allergic reactions may develop.

Overdose

There are no data on cases of drug overdose.

With prolonged use of Pancreoflat in high doses, it is possible to increase the level of uric acid in the blood plasma and develop hyperuricosuria.

special instructions

Due to the lack of information on the toxic effects of pancreatin on reproductive function or fetal development, pregnant and breastfeeding women should not start using the drug without consulting a doctor.

Influence on the ability to drive vehicles and complex mechanisms

The use of the drug does not affect the psychomotor functions associated with driving vehicles and working with complex mechanisms.

Application during pregnancy and lactation

There are no data on the toxic effect of the drug when used during pregnancy and lactation, but since the safety of pancreatin and dimethicone has not been studied enough, Pancreoflat should be used with caution in pregnant and breastfeeding women.

Pediatric use

Pancreoflat tablets are contraindicated in children under 12 years of age.

Drug interactions

With concomitant therapy with antacids containing aluminum hydroxide and / or magnesium carbonate, the therapeutic effect of dimethicone may decrease.

With the simultaneous use of Pancreoflat with other drugs, no clinically significant interactions were noted.

Reviews about Pancreoflat

Reviews about Pancreoflat are controversial. In positive messages, patients describe the high efficacy of the drug and note that it reduced flatulence and significantly improved digestion. However, they often complain about the lack of this enzymatic agent in pharmacies.

In negative comments, patients write that the therapy did not help eliminate the existing disorders of the digestive tract. Doctors, however, draw attention to the fact that Pancreoflat is not used to treat diseases, it only replaces the missing pancreatic enzymes and prevents the formation of gas bubbles in the intestines, so it is important to find out the cause of the problem.
