Release form and composition
Dosage form - film-coated tablets: round, biconvex, rough, from white to white with a cream shade, at a break from white to white with a cream or greenish-yellowish shade of color (10 pcs. In cell contour packs, in cardboard pack of 1, 2 or 4 packages and instructions for use of Afludol).
Composition of 1 tablet:
- active substance: umifenovir hydrochloride (in the form of monohydrate) - 50 or 100 mg;
- auxiliary components: crospovidone, povidone K-25 (medium molecular weight polyvinylpyrrolidone), magnesium stearate, colloidal silicon dioxide (aerosil), lactose monohydrate;
- film shell: opadry II white, including macrogol-4000 (polyethylene glycol 4000), hypromellose, lactose monohydrate, titanium dioxide.
Pharmacological properties
Pharmacodynamics
The active ingredient of Afludol, umifenovir, is an antiviral agent. Specifically inhibits influenza A and B viruses and coronavirus associated with severe acute respiratory syndrome (SARS).
According to the mechanism of antiviral action, umifenovir belongs to fusion inhibitors: it interacts with the hemagglutinin of the virus and prevents the fusion of its lipid membrane with cell membranes.
The drug has a moderate immunomodulatory activity and the ability to induce interferons. Stimulates the phagocytic function of macrophages, cellular and humoral immune responses. Increases the body's resistance to viral infections. Reduces the frequency of exacerbations of chronic bacterial diseases, as well as the development of complications associated with a viral infection.
In viral infections, the therapeutic efficacy of umifenovir is manifested in weakening the severity of clinical symptoms and general intoxication of the body, in reducing the duration of the disease and reducing the risk of complications.
Umifenovir belongs to low-toxic drugs [LD50 (half-lethal dose) is more than 4 g / kg]. When taken in recommended doses, it does not have a negative effect on the human body.
Pharmacokinetics
After oral administration, umifenovir is rapidly absorbed and distributed to tissues and organs. When used in a dose of 50 mg, the maximum concentration reaches after 1.2 hours, in a dose of 100 mg - after 1.5 hours.
It is metabolized in the liver. The half-life (T 1/2) is 17-21 hours.
Approximately 40% of the dose received is excreted unchanged: 38.9% - with bile, 0.12% - by the kidneys. In this case, up to 90% of the dose taken is excreted during the first day.
Indications for use
- treatment and prevention of influenza A and B, as well as other acute respiratory viral infections (ARVI);
- complex therapy of pneumonia, chronic bronchitis, recurrent herpes infection;
- prevention of postoperative infectious complications.
Contraindications
- glucose-galactose malabsorption, lactose intolerance, lactase deficiency;
- children under 3 years old;
- hypersensitivity to any component of the drug.
Afludol, instructions for use: method and dosage
Afludol tablets should be taken orally, before meals.
Recommended single doses of the drug, depending on the patient's age:
- adults and adolescents from 12 years old - 200 mg (2 tablets of 100 mg or 4 tablets of 50 mg);
- children 6–12 years old - 100 mg (1 tablet 100 mg or 2 tablets 50 mg each);
- children 3-6 years old - 50 mg (1 tablet 50 mg).
Recommended schemes for taking Afludol, depending on the indications:
- non-specific prevention of acute respiratory viral infections and influenza during an epidemic: 1 dose 2 times a week for 3 weeks;
- non-specific prevention of acute respiratory viral infections and influenza after direct contact with the patient: 1 dose 1 time per day for 10-14 days;
- treatment of influenza and ARVI: 1 dose 4 times a day (at intervals of 6 hours) for a course of 5 days;
- complex therapy of pneumonia, chronic bronchitis, recurrent herpetic infection: 1 dose 4 times a day (at intervals of 6 hours) for 5-7 days, then - 1 dose 2 times a week for another 4 weeks;
- prevention of postoperative infectious complications: 1 dose 2 days before surgery, then 1 dose on days 2 and 5 after surgery.
Side effects
Afludol is generally well tolerated. In rare cases, it can cause allergic reactions.
Overdose
To date, no cases of overdose have been reported.

Afludol has no central neurotropic activity, therefore it can be used for prophylaxis in practically healthy people employed in various fields of activity, including those requiring good coordination of movements and increased concentration of attention.
Pediatric use
Afludol tablets are contraindicated in children under 3 years of age.
