Semax is a nootropic drug with a pronounced antioxidant, cerebroprotective and antihypoxic effect.

Release form and composition

Semax is produced in the form of nasal drops of 0.1% and 1%: transparent, colorless (3 ml in vials, in a cardboard box 1 vial complete with a pipette stopper).

Active ingredient: methionyl-glutamyl-histidyl-phenylalanyl-prolyl-glycyl-proline, in 1 ml drops - 1 or 10 mg.

Excipients: methyl parahydroxybenzoate, purified water.

Pharmacological properties

Semax is a synthetic peptide drug, the active component of which is an analogue of a fragment of adrenocorticotropic hormone (ACTH 4-10) methionyl-glutamyl-histidyl-phenylalanyl-prolyl-glycyl-proline. This substance completely lacks hormonal activity. All amino acids included in it belong to the L-form.

Methionyl-glutamyl-histidyl-phenylalanyl-prolyl-glycyl-proline is distinguished by an original mechanism of neurospecific action on the central nervous system. Semax is a synthetic analogue of corticotropin, characterized by nootropic properties and practically not showing hormonal activity. The drug affects the processes associated with learning and memory formation. Semax promotes concentration of attention during training and information collection, ensures the adaptation of the body to anesthesia, cerebral ischemia, hypoxia and other traumatic influences, improves the consolidation of the memory footprint in patients with cerebrovascular diseases (including discirculatory encephalopathy), as well as after traumatic brain injury and neurosurgical interventions.

The drug does not exhibit toxic properties both with single and multiple administration. Also, mutagenic, teratogenic, embryotoxic and allergic properties are uncharacteristic for it. Methionyl-glutamyl-histidyl-phenylalanyl-prolyl-glycyl-proline does not have a local irritating effect.

Pharmacokinetics

The absorption of methionyl-glutamyl-histidyl-phenylalanyl-prolyl-glycyl-proline occurs from the nasal mucosa, and up to 60–70% of the substance is fully absorbed. It is distributed at a high speed to all organs and tissues, and also penetrates the blood-brain barrier. When it enters the bloodstream, the drug undergoes rapid biotransformation and is excreted from the body through the kidneys.

Indications for use

Nasal drops 0.1%:

  • increasing the adaptive capabilities of the human body in extreme situations;
  • transient disorders of cerebral circulation (transient ischemic attack);
  • encephalopathy;
  • neurotic disorders of various origins, including after ionizing radiation;
  • the recovery period after a stroke;
  • conditions after anesthesia, craniocerebral trauma, neurosurgical interventions;
  • intellectual and mnestic disorders due to vascular lesions of the brain;
  • atrophy of the optic nerve, neuritis of toxic-allergic and inflammatory etiology;
  • minimal brain dysfunction (neuropsychiatric disorders, including attention deficit hyperactivity disorder) in children from 7 years old.

Also, in the form of 0.1% nasal drops, Semax is recommended for people whose work is associated with the performance of monotonous operator activities (for the prevention of mental fatigue during the most intense or stressful periods of work).

Nasal drops of 1% are prescribed for moderate and severe ischemic stroke, in the acute period of the disease (as part of combination therapy).

Contraindications

  • acute mental conditions;

Instructions for the use of Semax: method and dosage

Semax drops are used intranasally.

Method of using the dropper bottle:

  1. cut off the tip of the pipette with scissors;
  2. turn the bottle over and gently tap the bottom with your finger until the liquid fills the entire space of the pipette;
  3. remove the cap;
  4. squeeze out the required number of drops into the nasal passage, pressing on the wide part of the pipette.

Nasal drops 0.1%

1 drop of solution (using a pipette) contains 50 μg of active substance. No more than 2-3 drops can be injected into each nasal passage at a time. If a higher dose is prescribed, the administration should be carried out in several doses at intervals of 10-15 minutes.

Recommended dosage regimens depending on indications:

  • intellectual and mnestic disorders in vascular lesions of the brain, transient disorders of cerebral circulation, discirculatory encephalopathy: a single dose - 3-30 mcg / kg (200-2000 mcg), daily dose - 7-70 mcg / kg (500-5000 mcg), frequency application - up to 4 times a day, duration of treatment - 10-14 days, if necessary, appoint repeated courses;
  • conditions after neurosurgical interventions, craniocerebral trauma, anesthesia: 40-50 mcg / kg (1400-3500 mcg) 3 times a day for 3-5 days, if necessary, the duration of therapy is increased to 14 days;
  • neuritis and optic nerve atrophy: 100-150 mcg in each nasal passage 2-3 times a day, daily dose - 600-900 mcg, duration of treatment is 7-10 days. If necessary, the drug is administered by endonasal electrophoresis - from the anode, using a current of 1 mA. The daily dose is 400-600 mcg. The duration of exposure is 8-12-15 minutes. The course of treatment is 7-10 days.

To prevent mental fatigue and increase the adaptive capabilities of the human body, it is recommended to use Semax 2-3 drops in each nasal passage 2-3 times in the morning. The daily dose ranges from 400 to 900 mcg. The duration of therapy is 3-5 days, if necessary, repeated courses are carried out.

With minimal cerebral dysfunction, children from 7 years old are prescribed 1-2 drops of the drug (50-100 mcg or 5-6 mcg / kg) in each nasal passage 2 times a day - in the morning and in the evening. The maximum daily dose is 400 mcg. The duration of treatment is 30 days.

Nasal drops 1%

1 drop of solution (when using a pipette) contains 500 μg of active substance.

Recommended dosages:

  • stroke of moderate severity: 2-3 drops in each nostril (2000-3000 mcg) 3-4 times a day at 3-4-hour intervals. The daily dose is 6000-12000 mcg (12-24 drops);
  • severe stroke: 3-4 drops in each nasal passage (3000-4000 mcg) 4-5 times a day at 2.5-3-hour intervals. The daily dose is 12,000-20,000 mcg (24-40 drops).

The duration of treatment is 10 days.

Side effects

According to the instructions, Semax is usually well tolerated. With long-term treatment, slight irritation of the nasal mucosa is possible.
