Release form and composition
The dosage form of the drug Ornidazole-vero is film-coated tablets: biconvex, round, from white to white with a yellowish tinge, two layers are noticeable in the cross section (in a cell contour packaging or a polymer can 10 pcs., In a cardboard box 1 package or bank).
Composition of one tablet:
- active substance: ornidazole - 500 mg;
- auxiliary components (core): microcrystalline cellulose, corn starch, polyvinylpyrrolidone (povidone), primogel (sodium carboxymethyl starch), magnesium stearate;
- shell: hydroxypropyl methylcellulose (hypromellose), polyvinylpyrrolidone (povidone), polysorbate 80 (tween-80), talc, titanium dioxide.
Pharmacological properties
Pharmacodynamics
Ornidazole belongs to antiprotozoal agents derived from 5-nitroimidazole. It has a bactericidal (death of microorganisms) and bacteriostatic (stopping the growth of bacteria) action. By biochemical reduction of the 5-nitro group of ornidazole by transport intracellular proteins of anaerobic microorganisms, further interaction of the reduced 5-nitro group with deoxyribonucleic acid (DNA) of bacteria and inhibition of the synthesis of their nucleic acids, death of anaerobic microbes and protozoa occurs. In addition, the active substance has a toxic effect on microorganisms, inhibiting the process of their cellular respiration.
Ornidazole is active against Giardia lamblia, Gardnerella vaginalis, Trichomonas vaginalis, Entamoeba histolytica, gram-negative anaerobic rod-shaped bacteria (including Bacteroides ovatus, Bacteroides distasonis, Bacteroides fragilisgatus, Bacteroides vulgaris)., Fusobacterium spp., And Prevotella spp. (including Prevotella disiens, Prevotella buccae, Prevotella bivia).
Aerobic bacteria are not sensitive to ornidazole.
Pharmacokinetics
- absorption: well absorbed in the gastrointestinal tract. Bioavailability is 90%, the maximum concentration in blood plasma is reached after 1-2 hours;

Indications for use
- giardiasis;
- amoebic dysentery, extraintestinal forms of amebiasis (including amoebic abscess of the liver, lungs, skin amebiasis);
- trichomoniasis;
- prevention of postoperative infections caused by anaerobic microorganisms, during surgery on the colon and in gynecology.
Contraindications
Absolute:
- age up to 3 years;
- increased individual sensitivity to ornidazole, other nitroimidazole derivatives or any of the auxiliary components of the drug.
Relative (the use of the drug Ornidazole-vero requires caution):
- organic diseases of the central nervous system (including multiple sclerosis, epilepsy);
- liver disease;
- pregnancy, lactation period;
- alcoholism.
Instructions for use of Ornidazole-vero: method and dosage
Ornidazole-vero 500 mg tablets are taken orally, after meals.
The dosage regimen depends on the disease, age and weight of the patient.
Giardiasis
- adults and children weighing over 35 kg: 3 tablets (1500 mg) once a day, the course of treatment is 1-2 days;
- children over 3 years old weighing less than 35 kg: at the rate of 40 mg / kg body weight 1 time per day for 1-2 days.
Amebic dysentery
- adults and children weighing 35 kg or more: 3 tablets (1500 mg) once a day, the course of treatment is 3 days;
- adults weighing more than 60 kg: 4 tablets (2000 mg) once a day for 3 days;

Extraintestinal forms of amebiasis
- adults and children weighing over 35 kg: 1 tablet (500 mg) 2 times a day for 5-10 days;

Trichomoniasis
- adults: 1 tablet (500 mg) 2 times a day for 5 days;

Prevention of postoperative infections
It is recommended to take one tablet (500 mg) before surgery, then 1 tablet every 12 hours for 3-5 days.
Side effects
- from the digestive system: nausea, vomiting, diarrhea, perversion of taste, impaired activity of liver enzymes;
- from the central and peripheral nervous system: dizziness, headache, drowsiness, tremors, temporary loss of consciousness, muscle stiffness, impaired coordination of movements, convulsions, sensory or mixed variants of peripheral neuropathy.
Overdose
In case of an overdose of Ornidazole-vero, symptoms such as depression, epileptiform seizures, and peripheral neuritis may occur.

Pediatric use

Drug interactions
When used simultaneously with coumarin anticoagulants, ornidazole can enhance their effect, therefore, in such cases, an appropriate dose adjustment of anticoagulants is required.
Ornidazole-vero prolongs the muscle relaxant effect of vecuronium bromide.
Since ornidazole does not suppress acetaldehyde dehydrogenase in the liver, it is compatible with alcohol, unlike other imidazole derivatives.
