Lotseril is an antifungal drug for external use.

Release form and composition

Lotseril is available in the form of 5% nail polish: an almost colorless or colorless transparent liquid (1.25 ml, 2.5 ml or 5 ml each in dark glass vials with a plastic cap with a built-in applicator and a polymer insert, 1 bottle in a cardboard box complete with 30 nail files and 30 sealed envelopes with isopropyl alcohol swabs).

1 ml of varnish contains:

  • active substance: amorolfine hydrochloride - 64 mg, this corresponds to the content of 57.4 mg of amorolfine base;
  • auxiliary components: triacetin, copolymer of methyl methacrylate, trimethylammonioethyl methacrylate chloride, ethyl acrylate (1: 2: 0.2), butyl acetate, absolute ethanol, ethyl acetate.

Pharmacological properties

Pharmacodynamics

Lotseril is a drug for external use with a wide spectrum of antifungal action (fungicidal and fungistatic). The active substance is amorolfine, which has the ability to disrupt sterol biosynthesis. A decrease in ergosterol levels causes the accumulation of atypical steric non-planar sterols, resulting in damage to the cytoplasmic membrane of the fungus.

Possesses high activity against common and rare pathogens of fungal nail infections.

The following microorganisms show sensitivity to the drug:

  • dermatophytes: Trichophyton speciales (spp.), Epidermophyton spp., Microsporum spp.;
  • molds: Alternaria spp., Hendersonula spp., Scopulariopsis spp.;
  • yeast fungi: Cryptococcus spp., Candida spp., Pityrosporum spp. (Malassezia spp.);
  • dimorphic fungi: Histoplasma spp., Coccidioides spp., Sporothrix spp.;
  • mushrooms from the family Dematiaceae: Fonsecaea spp., Cladosporium spp., Wangiella spp.

Pharmacokinetics

After applying Lotseril, the penetration of varnish into the nail plate and the nail bed occurs almost completely within the first 24 hours. After the first application, the therapeutically effective concentration of amorolfine is maintained for 7-10 days.

Due to the insignificant systemic absorption, the plasma content of the drug is practically undetectable.

Indications for use

  • onychomycosis caused by dermatophytes, yeasts and molds;
  • prevention of fungal infections on the nails.

Contraindications

  • childhood;
  • period of pregnancy;
  • breast-feeding;
  • hypersensitivity to the components of the drug.

Instructions for the use of Lotseril: method and dosage

Loceryl varnish is intended for external use by applying to the nails of the fingers or toes affected by the fungus.

Before each application of the drug, the surface of the nail plate is thoroughly cleaned with the attached file, removing already destroyed tissues of the affected area of the nail. Then, using a tampon soaked in alcohol, the surface of the nail is degreased.

Having lowered the applicator into the bottle, carefully remove it without touching the walls of the bottle neck, and apply varnish on the surface of the entire affected nail in an even layer.

The procedure is similarly carried out for each affected nail. The varnish dries up within three minutes.

Close the bottle tightly after each use. Wipe the applicator with the swab that was used to clean the nail, avoiding contact of the alcohol swab with the treated fingernails.

After the procedure, you must thoroughly wash your hands, waiting for the varnish to dry completely.

The recommended frequency of varnish application is 1-2 times a week.

The use of Lotseril varnish should be continued until the affected area is completely healed and the nail regenerates. For fingernails, the average duration of treatment is 24 weeks, for toenails - 36–48 weeks.

The first signs of a therapeutic effect can be observed only after 8-12 weeks of therapy.

If, after 12 weeks of regular application of Lotseril, there are no signs of improvement, you should consult a doctor.

Side effects

  • on the part of the immune system: the frequency has not been established - hypersensitivity reactions, allergic reactions of a systemic nature;
  • dermatological reactions: rarely - discoloration of nails, damage to nails, onychoclasia, fragile structure of the nail plate; very rarely - burning sensation of the skin; frequency not established - pruritus, erythema, urticaria, contact dermatitis, blistering.

Overdose

Symptoms of a systemic overdose are unlikely if the recommendations for the use of the drug are followed.

Treatment: if the varnish accidentally gets inside, it is necessary to wash the stomach, then symptomatic therapy is indicated.

If more than 2/3 of the nail plate is affected by a fungal infection, it is recommended to consider the appointment of concomitant oral antifungal therapy.

The development of undesirable reactions against the background of the use of varnish is rarely noted. The observed destruction and discoloration of the nail plates or brittle nails may be due to onychomycosis. In the event of local or systemic allergic reactions, the use of Lotseril should be stopped immediately and consult a doctor.

It is allowed to use soap with regular washing of hands or feet.

You can apply decorative nail polish 10 minutes after Lotseril. Before the next procedure, the decorative varnish must be carefully removed, the affected areas of the nails must be treated with a nail file, and the remaining layer of the drug must be wiped off with an alcohol swab.

Files used to treat affected nails should not be used on healthy nails.

The content of ethanol in the composition of Lotseril, with too frequent or incorrect application of varnish, can cause irritation or dryness of the skin around the nail.

Avoid getting the varnish on mucous membranes, eyes or ears. If varnish accidentally gets into your eyes, rinse them immediately with running water.

The use of Lotseril for the prevention of the development of fungal infections of the nails is indicated for patients at risk. Conditions that increase the risk of developing onychomycosis include: diabetes mellitus, impaired peripheral circulation, immunodeficiency, chronic skin diseases (including psoriasis), nail degeneration, destruction of the nail plate.

The swab is impregnated with a highly flammable substance.

Application during pregnancy and lactation

According to the instructions, Lotseril is not recommended for use during gestation and breastfeeding.

Pediatric use
