Andriol is a drug that compensates for the deficiency of testosterone, an endogenous hormone that is necessary for the normal development of male genital organs and secondary sexual characteristics.

Release form and composition

Dosage form - capsules with a dosage of 40 mg.

The main active ingredient is testosterone undecanoate.

Excipients: medium chain triglycerides, propylene glycol monolaurate, lecithin, glycerol, castor oil, gelatin, dye.

Pharmacodynamics and pharmacokinetics

The use of Andriol in men with hypogonadism, with an individual dose selection, normalizes the total concentration of testosterone in the blood serum and increases the level of bioavailable testosterone. During therapy, the content of dihydrotestosterone and estradiol in the blood serum rises, and the level of globulin, which binds sex hormones, follicle-stimulating hormone and luteinizing hormone, decreases.

The active component of the drug - testosterone undecanoate - causes a lesser severity of symptoms of testosterone deficiency in both boys and adult men with hypogonadism. Its use leads to an increase in bone mineral density and lean body mass, and also improves sexual function, in particular potency and libido.

Depending on the dose of testosterone undecanoate, a decrease in the concentration of low-density lipoproteins, high-density lipoproteins and triglycerides, an increase in hemoglobin and hematocrit, clinically insignificant changes in the content of liver enzymes and prostate specific antigen are observed. Treatment can cause an increase in the size of the prostate, but there are no side effects from the prostate. Patients with hypogonadism and diabetes have increased insulin sensitivity and decreased blood glucose levels.

The use of Andriol in boys with delayed growth and puberty accelerates the growth and development of secondary sexual characteristics.

A single dose of the drug in a dose of 80-160 mg causes a clinically significant increase in the total concentration of testosterone in plasma. Its maximum level of approximately 40 nmol / L is reached 4–5 hours after ingestion. The increased testosterone content in the blood plasma persists for at least 8 hours. Testosterone undecanoate and testosterone bind almost 97% non-specifically to SHBG and blood proteins.

In tissues and plasma, the active substance Andriol is hydrolyzed to the natural male androgen - testosterone, which is further metabolized to estradiol and dihydrotestosterone.

Testosterone and its metabolites are excreted mainly in the urine in the form of conjugates (androsterone and etiocholanolone).

Indications for use

Andriol is prescribed as hormone replacement therapy for diseases and conditions caused by a lack of testosterone:

  • Endocrine impotence;
  • Eunuchoidism;
  • Post-castration syndrome;
  • Hypopituitarism;
  • Infertility associated with impaired spermatogenesis;
  • Menopause in men (decreased libido, deterioration in general health);

Contraindications

The use of the drug is contraindicated in case of suspicion of breast or prostate carcinoma, as well as in case of hypersensitivity to the components.

The initial dose is 120-160 mg / day for 2-3 weeks. For maintenance therapy, the drug is taken at 40-120 mg / day. The daily dose is divided into two doses.

  • insulin and other hypoglycemic drugs: an increase in glucose tolerance and a decrease in the need for insulin or other hypoglycemic drugs in patients with diabetes mellitus (special monitoring is recommended before, during and after treatment with Andriol);
  • coumarin-type anticoagulants: increasing the effect of the latter while taking high doses of testosterone undecanoate (prothrombin time should be carefully monitored and, if necessary, the dose of anticoagulant should be reduced).

According to the instructions, Andriol is able to reduce the content of thyroxine-binding globulin, as a result of which the level of total T4 in serum decreases and the binding of T3 and T4 increases. At the same time, the level of free thyroid hormones does not change, and there are no clinical symptoms of thyroid dysfunction.
