Pharmacological properties

Pharmacodynamics

Sulfacetamide is an antimicrobial drug with a bacteriostatic effect for topical use in ophthalmology. Its active substance is sulfacetamide, an intermediate product of the synthesis of streptocide, belongs to the pharmacological group of sulfonamides, and is a competitive antagonist of para-aminobenzoic acid. The mechanism of action of the drug is due to such properties of sulfonamides as the ability to inhibit dihydropteroate synthetase and disrupt the synthesis of tetrahydrofolic acid, which is involved in the formation of purines and pyrimidines.

Sulfacetamide is sensitive to gram-positive and gram-negative cocci, Shigella spp. (speciales), Chlamydia spp., Vibrio cholerae, Escherichia coli, Corynebacterium diphtheriae, Clostridium perfringens, Yersinia pestis, Toxoplasma gondii, Actinomyces israelii, Bacillus anthracis.

Pharmacokinetics

After application, the maximum concentration (C max) of sulfacetamide is reached in the first 0.5 hours. In this case, C max in the cornea is about 3 mg / ml, the moisture of the anterior chamber is ~ 0.5 mg / ml, the iris is ~ 0.1 mg / ml … In the tissues of the eyeball, the drug is stored for 3-4 hours at a concentration of less than 0.5 mg / ml.

With damage to the corneal epithelium, the penetrating activity of sulfonamides increases.

Indications for use

The use of eye drops Sulfacetamide is indicated in the complex therapy of the following diseases:

  • conjunctivitis;
  • purulent corneal ulcer;
  • blepharitis;
  • gonorrheal and chlamydial eye infections.

In addition, in newborns, drops are used for the prevention and treatment of blennorrhea.

Contraindications

It is contraindicated to use drops with established hypersensitivity to the components that make up their composition.

With caution, only on the recommendation of a doctor, the use of sulfonamides during pregnancy and lactation is allowed.

Sulfacetamide, instructions for use: method and dosage

Sulfacetamide eye drops are applied topically, by instillation into the conjunctival sac of each eye.

Recommended dosage:

  • conjunctivitis in adults and children: 1-2 drops every 2-3 hours for 7-10 days;
  • blepharitis in adults and children: 1-2 drops 6-8 times a day in the acute period of the disease, as the condition improves, the frequency of instillations decreases, and can be from 3 to 4 times a day. The drug should be used for 3-5 days;
  • purulent corneal ulcer (as part of complex therapy): 1-2 drops 5-6 times a day. The duration of the course of treatment is 5–7 days;
  • gonorrheal and chlamydial eye diseases (as part of complex therapy in adults): 1–2 drops 5–6 times a day for 28–42 days. With positive dynamics, a decrease in the frequency of procedures with an interval of 7 days is shown;
  • prevention of blenorrhea in newborns: immediately after birth - 2 drops in each conjunctival sac, then after 2 hours the procedure must be repeated.

Side effects

  • on the part of the organ of vision: burning, itching and / or pain in the eyes, transient blurred vision after instillation, lacrimation;
  • from the immune system: allergic reactions.

Overdose

Overdose symptoms have not been established.

special instructions

It should be borne in mind that hypersensitivity to Sulfacetamide can be observed in patients with established individual intolerance to thiazide diuretics, furosemide, sulfonylurea or carbonic anhydrase inhibitors. This category of patients is recommended to use the drug with caution.

Influence on the ability to drive vehicles and complex mechanisms

If, against the background of the use of the drug, a temporary violation of the clarity of vision occurs, then immediately after instillation, patients are not recommended to drive vehicles or engage in those activities that require clarity of visual perception.

Application during pregnancy and lactation

Sulfacetamide should be used during pregnancy or lactation only in cases where, in the opinion of the doctor, the expected clinical effect for the mother outweighs the potential risk to the fetus or child.

Pediatric use

Sulfacetamide in childhood should be used only according to indications and in doses recommended for treatment and prevention.

Drug interactions

It should be borne in mind that Sulfacetamide is pharmacologically incompatible with silver salts.

With simultaneous use with procaine or tetracaine, the bacteriostatic effect of drops is reduced.

Analogs

The analogues of Sulfacetamide are Albucid, Sulfacil sodium, Sulfacil sodium Vial, Sulfacil sodium-DIA, Sulfacil sodium-SOLOpharm.
