Siflox is a broad spectrum antibacterial agent.

Release form and composition

Dosage form of Siflox - coated tablets (10 pcs. In a blister, 1 blister in a cardboard box).

The drug contains the active substance ciprofloxacin (in the form of monohydrate hydrochloride), in 1 tablet - 250 or 500 mg.

Auxiliary components: microcrystalline cellulose, corn starch, colloidal silicon dioxide, magnesium stearate, crospovidone; film casing - Opadry White OY S 28842.

Pharmacological properties

Pharmacodynamics

Ciprofloxacin is a broad-spectrum antimicrobial agent. Refers to the group of fluoroquinolones. Possesses bactericidal properties. The mechanism of action is due to the ability to suppress DNA gyrase and inhibit the synthesis of bacterial DNA.

Shows high activity against most gram-negative bacteria, including Haemophilus influenzae, Neisseria meningitidis, Shigella spp., Escherichia coli, Pseudomonas aeruginosa, Neisseria gonorrhoeae, Salmonella spp.

Effective against Staphylococcus spp. (including methicillin-resistant strains and strains producing and not producing penicillinase), some strains of Enterococcus spp., Mycoplasma spp., Mycobacterium spp., Legionella spp., Campylobacter spp., Chlamydia spp., as well as bacteria producing beta-lactamases …

Microorganisms Clostridium difficile, Ureaplasma urealyticum and Nocardia asteroides are resistant to the action of ciprofloxacin.

The effect of the drug against Treponema pallidum has not been sufficiently studied.

Pharmacokinetics

After oral administration, ciprofloxacin is rapidly absorbed from the gastrointestinal tract. Simultaneous food intake has a slight effect on absorption. Bioavailability is approximately 70%.

When taken on an empty stomach, the maximum serum concentration is reached within 30-180 minutes. Depending on the dose taken, the maximum concentration is:

  • 250 mg - 0.76 mg / ml;
  • 500 mg - 1.6 mg / ml;
  • 750 mg - 2.5 mg / ml;
  • 1000 mg - 3.4 mg / ml.

It binds to plasma proteins by 20-40%. Distributed in tissues and body fluids. Reaches high concentrations in bile. Penetrates into the cerebrospinal fluid: with non-inflamed meninges, the concentration of ciprofloxacin reaches 10%, with inflamed meninges - up to 37%.

Excreted in urine and bile.

Indications for use

According to the instructions, Siflox is used to treat infectious and inflammatory diseases caused by microorganisms sensitive to its active substance, including diseases of the respiratory tract and ENT organs, infections of the gastrointestinal tract, biliary tract, pelvic organs, genital organs (including gonorrhea), skin and soft tissues, bones and joints, eye infections, wound infections, postoperative septicemia.

Also Siflox is prescribed for the treatment and prevention of infections in patients with reduced immunity.

Contraindications

  • childhood;
  • pregnancy and lactation;
  • hypersensitivity to the components of Siflox or other drugs of the quinolone series.

With caution, Siflox should be used in the elderly, with disorders of cerebral circulation, atherosclerosis of the cerebral vessels, convulsive syndrome of unclear etiology, epilepsy.

Siflox tablets are taken orally.

Average effective doses depending on indications:

  • urinary tract infections: uncomplicated - 250 mg 2 times a day, severe - 500 mg 2 times a day;
  • severe respiratory tract infections, osteomyelitis: 750 mg 2 times a day;
  • other infections: 500 mg 2 times a day.

Duration of treatment:

  • urinary tract and kidney infections - 7-10 days;
  • streptococcal infections - at least 10 days.

Usually it is recommended to continue taking Siflox for 72 hours after clinical and microbiological recovery.

In acute gonorrhea, the drug is taken once in a dose of 250 mg.

For patients with severe renal impairment (creatinine clearance <20 mg / min), the usual dose is reduced by 2 times.

Side effects

  • from the digestive system: abdominal pain, nausea, diarrhea, vomiting, pseudomembranous colitis, increased bilirubin, alkaline phosphatase, hepatic transaminase activity, lactate dehydrogenase;
  • from the urinary system: polyuria, dysuria, hematuria, albuminuria, crystalluria, transient increase in serum creatinine, glomerulonephritis;
  • on the part of the hematopoietic system: changes in the number of platelets, leukopenia, eosinophilia, neutropenia;
  • from the central nervous system: fatigue, headache, visual disturbances, dizziness, hallucinations, sleep disorders, nightmares, fainting;

Siflox can increase the concentration of theophylline in blood plasma and increase its half-life, resulting in the development of toxic effects.

When used simultaneously with warfarin, ciprofloxacin increases the risk of bleeding.
