Kolofort is a drug with anti-inflammatory, antispasmodic and anxiolytic effects.
Release form and composition
The drug is produced in the form of tablets for resorption: flat-cylindrical, almost white or white, with a risk and a chamfer; on the side with a line the inscription "MATERIA MEDICA" is applied, on the other side - "KOLOFORT" (in a cardboard box 1, 2 or 5 cell contour packs of 20 tablets each and instructions for the use of Kolofort).
Composition of 1 tablet:
- active substances: affinity purified antibodies * to histamine - 0.006 g, affinity purified antibodies * to brain-specific protein S-100 - 0.006 g, affinity purified antibodies * to human tumor necrosis factor alpha (TNF-α) - 0.006 g;
- auxiliary components: lactose monohydrate, magnesium stearate, microcrystalline cellulose.
* They are applied to lactose in the form of a mixture of three active aqueous-alcoholic dilutions of the substance, diluted 100 200, 100 30 and 100 12 times, respectively.

It has been experimentally established that the active components of Kolofort modify the activity of ligand-receptor interaction of endogenous regulators with the corresponding receptors: antibodies to histamine - to histamine H 4 receptors located in the gastrointestinal tract (GIT), antibodies to TNF-α - to the TNF-α receptor, antibodies to protein S-100 - to serotonin receptors and σ 1 receptors.
Due to the combination of three components, Kolofort has a complex effect on the peripheral and central links in the pathogenesis of functional intestinal disorders, including abdominal pain syndrome.
Properties of the active substances of the drug:
- antibodies to histamine have antispasmodic, anti-edematous and anti-inflammatory effects;
- antibodies to TNF-α have a pronounced anti-inflammatory effect, help to normalize the balance of pro-inflammatory and anti-inflammatory cytokines;
- antibodies to the S-100 protein have a wide spectrum of psychotropic activity, including anxiolytic, antidepressant, nootropic and antiasthenic effects, which in therapeutic practice is manifested by the elimination of anxiety and internal tension, as well as the normalization of a number of visceral functions, including the activity of the large intestine. They are not sedative, addictive and non-withdrawal.
Due to the properties of active ingredients, Kolofort normalizes nervous and humoral regulation of intestinal function, reduces visceral hypersensitivity of colon receptors to stretching, reduces the severity of pain syndrome, relieves the feeling of stomach overflow and bloating, and provides restoration of impaired gastrointestinal motility.
The antispasmodic effect of the drug is expressed in relaxation of smooth muscles and a decrease in the tone of the gastrointestinal tract wall, a decrease in intraluminal pressure, normalization of stool frequency, its consistency and accompanying symptoms (relief of the feeling of incomplete bowel movement, tenesmus, imperative urge, additional efforts during the act of defecation, etc.).

The study of the pharmacokinetics of Kolofort is technically impossible, since biological fluids, tissues and organs contain ultra-low doses of antibodies that are not determined by modern physicochemical methods of analysis, such as chromatography-mass spectrometry, gas-liquid chromatography and high-performance liquid chromatography.

Contraindications
Kolofort tablets are contraindicated in case of increased individual sensitivity to their components.

The drug is taken orally between meals. The tablets should be kept in the mouth until they are completely dissolved.
The recommended dose is 1-2 tablets 2 times a day. During an exacerbation of the disease, a temporary (for 7-14 days) increase in the frequency of taking the drug up to 4 times a day is possible.
The duration of treatment is 1–3 months. If necessary, the course is extended up to 6 months and / or repeated after 1–2 months.
Side effects
Against the background of the use of Kolofort, the development of reactions of individual hypersensitivity to the components of the drug is possible.
Overdose
In case of overdose, dyspeptic disorders may occur.
