KONDRONOVA is a combined chondrostimulating drug that regulates the exchange of cartilage tissue.

Release form and composition

Capsules: size No. 0, hard, gelatinous, blue; content - white granular powder (10 pcs. in blisters, 3 or 6 blisters in a cardboard box).

Composition of 1 capsule:

  • active substances: glucosamine sulfate - 250 mg; chondroitin sulfate - 200 mg;
  • additional components: magnesium stearate, croscarmellose sodium, mannitol, purified talc;
  • capsule shell: sodium lauryl sulfate, bronopol, gelatin, purified water, titanium dioxide, povidone, quinoline yellow and brilliant blue dyes.

Ointment: light brown (10, 30 or 50 g in tubes; 1 tube in a cardboard box).

Composition of 1 g of ointment:

  • active substances: glucosamine sulfate - 25 mg, chondroitin sulfate - 50 mg;
  • additional components: mineral oil, white beeswax, white soft paraffin, lanolin (anhydrous), hydroxytoluene butylate, propylparaben, methylparaben.

Pharmacological properties

Pharmacodynamics

The active substances of the Kondronova preparation take part in the biosynthesis of the connective tissue, thereby preventing the processes of cartilage destruction and stimulating tissue regeneration. In addition, the drug has anesthetic and anti-inflammatory effects.

Chondroitin sulfate and glucosamine sulfate are amino saccharides that are the structural basis of articular cartilage and intervertebral disc cartilage. Namely, glycosaminoglycans (mucopolysaccharides) of the intercellular matrix of cartilage, providing elasticity and sliding of the articular surfaces, as well as carbohydrate groups of collagen fibers of cartilage tissue: type II (fibrillar), VI type (microfibrillar), IX type (main) and XI type (assembly), providing strength and elasticity of the cartilage.

Glucosamine sulfate is one of the components of the synthesis of hyaluronic acid, which is involved in the formation of proteoglycans of the structural matrix of joints. This substance provides nonspecific protection of cartilage from chemical damage. In the form of sulfate, glucosamine can serve as a precursor of hexosamine for articular cartilage, and sulfate ion is required for the synthesis of glycosaminoglycans, which are esters of sulfuric acid. Glucosamine protects damaged cartilage from metabolic destruction due to the use of glucocorticosteroids (GCS) or non-steroidal anti-inflammatory drugs (NSAIDs). When taken orally, the drug stimulates the production of cartilage matrix and has a moderate anti-inflammatory effect.

Chondroitin sulfate serves as an additional substrate for the formation of a healthy cartilage matrix. The substance itself and its derivatives stimulate the synthesis of type II collagen and proteoglycans, as well as promote the formation of hyaluronan and protect it from enzymatic breakdown and the damaging effect of free radicals. Also, chondroitin sulfate inhibits the activity of enzymes that break down cartilage (elastase, hyaluronidase), stimulates cartilage repair mechanisms and maintains the viscosity of the synovial fluid. In the treatment of osteoarthritis, the drug relieves negative symptoms and reduces the need for NSAIDs.

Pharmacokinetics

The oral bioavailability of glucosamine sulfate due to the effect of the first passage through the liver is 25%. After oral administration, radioactively labeled glucosamine is found first in plasma components and later penetrates into tissues. The highest concentrations were found in the articular cartilage, kidneys and liver. About 30% of the received dose persists for a long time in the tissues of bones and muscles.

Most of the glucosamine found in feces is the unabsorbed fraction. The fraction of glucosamine that does not bind to plasma proteins or is not metabolized is excreted mainly in the urine. The terminal half-life of glucosamine sulfate bound to plasma proteins is 68 hours.

After ingestion of chondroitin sulfate in a dose of 0.8 g once (or 0.4 g twice a day), the concentration of the substance in the plasma increases sharply within 24 hours. The absolute bioavailability of the drug is about 12%. Low molecular weight chondroitin sulfate is metabolized by desulfurization. It is excreted in the urine. The half-life is 310 minutes.

Indications for use

Kondronov capsules are used for the treatment of osteoarthritis of the peripheral joints and joints of the spine of I-III degree.

In the form of an ointment, the drug is recommended for use in the following cases:

  • osteoarthritis of the I – III degree;
  • deforming osteoarthritis;
  • osteoarthritis;
  • gonarthrosis;
  • spondylosis;
  • primary and secondary arthrosis;
  • scapular-humeral periarthritis;
  • chondromalacia of the patella;
  • intervertebral osteochondrosis.

Contraindications

  • bronchial asthma;

Instructions for the use of Kondronova: method and dosage

Kondronov capsules are taken orally. Recommended dosage: 2 pcs. 2 or 3 times a day for 2-3 months. After 60 days of therapy, the dose is gradually reduced depending on the individual needs of the patient. If necessary, a second course is carried out on the recommendation of a doctor.

The ointment is applied externally. Apply to intact skin over the affected area and rub in for 2-3 minutes. The course of treatment, depending on the indications and the severity of the disease, lasts from 2 weeks to 4 months.

Side effects

When Kondronova is used in capsule forms, mild gastrointestinal disorders (epigastric pain, constipation or diarrhea, flatulence), dizziness, headache, drowsiness or insomnia, tachycardia, allergic skin reactions, leg pain and peripheral edema are most often possible. In rare cases, there is an exacerbation of bronchial asthma and the development of more serious allergic reactions.

In the form of Kondronova ointment, it is mostly well tolerated, in some cases allergic reactions occur.

Overdose

Overdose cases have not been reported. If the dose of the drug in oral dosage form is significantly exceeded, gastric lavage and symptomatic therapy are recommended to eliminate the violations that have arisen.

When extremely high doses of glucosamine are taken orally (many times the recommended daily dose), hemorrhagic rashes, nausea or diarrhea may occur.
