Release form and composition

Lazolvan Rino dosage form is a dosed nasal spray: a clear solution of a pale yellow color with the smell of eucalyptus (10 ml in glass brown bottles equipped with a dispensing device and a nasal adapter with a protective cap, in a cardboard box 1 bottle).

Composition of 1 dose of the drug:

  • active substance: tramazoline hydrochloride monohydrate - 82 μg (in terms of tramazoline hydrochloride);
  • additional components: purified water - 66 358 mcg, povidone - 2101 mcg, glycerol 85% - 700 mcg, citric acid monohydrate - 270 mcg, sodium chloride - 183 mcg, sodium hydroxide - 154 mcg, magnesium sulfate heptahydrate - 49 mcg, hypromellose (hydroxypropyl methylcellulose) - 35 μg, magnesium chloride hexahydrate - 35 μg, L-menthol (levomenthol) - 14 μg, benzalkonium chloride - 14 μg, racemic camphor - 14 μg, calcium chloride dihydrate - 11 μg, cineole (eucalyptol) - 7 μg, sodium bicarbonate - 1 μg.

Pharmacological properties

Pharmacodynamics

Tramazoline hydrochloride - the active ingredient of Lazolvan Rino - is an alpha 2 -adrenomimetic that has a vasoconstrictor effect. Due to vasoconstriction after contact with the nasal mucous membranes, the drug reduces their swelling, as a result of which the patency of the nasal passages is quickly restored and nasal breathing is facilitated.

The drug begins to act 5 minutes after instillation, its effect lasts for 8-10 hours.

Pharmacokinetics

The pharmacokinetics of tramazoline in humans have not been studied. In the course of studies on monkeys, rabbits and rats, it was found that when the drug is administered orally or intranasally, from 50 to 80% of the dose received is absorbed. Both tramazoline and its metabolites are distributed in all internal organs; the highest concentration was found consistently in the liver.

The main metabolites of tramazoline after oral and topical administration are determined in urine.

The half-life is 5-7 hours.

Indications for use

According to the instructions, Lazolvan Rino is prescribed to relieve swelling of the mucous membrane and eliminate nasal congestion caused by acute respiratory diseases and / or hay fever (rhinitis, hay fever).

Contraindications

Absolute:

  • atrophic rhinitis;

Instructions for the use of Lazolvan Rino: method and dosage

Lazolvan Rino is administered intranasally.

Adults and children from 6 years old are prescribed 1 injection in each nostril. The frequency of instillations should not exceed 4 times a day. It is recommended to clear the nasal passages before injecting.

The duration of treatment is 5-7 days.

Use of the drug:

  1. Remove the protective cap from the bottle.
  2. Before the first use, press the dispenser several times into the air until a steady cloud of aerosol appears.
  3. Keeping the head straight, insert the tip into the nostril and press the dispenser once.
  4. Repeat the procedure for the second nasal passage.
  5. Remove the tip and inhale normally through the nose.
  6. Put a protective cap on the bottle.

It is recommended to clean the handpiece after each use.

  • from the skin and subcutaneous tissues (as symptoms of hypersensitivity): unknown frequency - rash, skin edema, itching;
  • from the nervous system: rarely - disturbances in taste, dizziness; infrequently - headache; unspecified frequency - sedation, drowsiness;
  • mental disorders: infrequently - anxiety; unknown frequency - insomnia, hallucinations.

Overdose

With an overdose of tramazoline, blood pressure (BP) may increase and tachycardia may occur, followed by them, especially in children, a sharp decrease in blood pressure, a decrease in body temperature, the development of reflex bradycardia and shock are possible.

Considering the experience of using other alpha-adrenergic agonists, the clinical picture of intoxication may be indistinct, which is due to the likelihood of a change in the phases of stimulation and depression of the central nervous system (CNS) and cardiovascular system (CVS), especially in children. CNS stimulation symptoms: agitation, anxiety, hallucinations, convulsions. Symptoms of CNS depression: lethargy, decrease in body temperature, drowsiness, coma.

Also, in case of an overdose, the following symptoms may develop: respiratory failure (including respiratory failure, respiratory arrest), cyanosis of the lips, miosis, mydriasis, pallor, fever, increased sweating, cardiac dysfunction (including bradycardia, cardiac arrest), mental disorders.

In case of a nasal overdose of tramazoline, it is necessary to urgently rinse the nose. When symptoms of intoxication appear, symptomatic therapy is performed.

The recommended duration of treatment should not be exceeded. If after 7 days there is no improvement in the condition, you need to consult a doctor who will decide whether it is advisable to continue therapy or the need to stop it.

Avoid getting the spray in the eyes.

With prolonged use, Lasolvan Rino, like other vasoconstrictor nasal agents, can cause the development of chronic nasal congestion and inflammation, as well as atrophy of the nasal mucosa.

Influence on the ability to drive vehicles and complex mechanisms

Special studies on the effect of tramazoline on the reaction rate and the ability to concentrate with its intranasal use have not been carried out. However, given the risk of such side effects from the central nervous system and psyche, such as sedation, drowsiness, fatigue, dizziness and hallucinations, it is recommended to refrain from performing potentially dangerous work and driving a car during the treatment period.

Application during pregnancy and lactation

According to long-term clinical experience of use, during pregnancy, Lazolvan Rino does not have a negative effect. During lactation, the safety of using the drug has not been confirmed.

In the first trimester of pregnancy, Lazolvan Reno should not be used. In the II and III trimesters, as well as during breastfeeding, the drug can be used, but only as directed by a doctor.

Pediatric use

In pediatrics, the drug is not used to treat patients under the age of 6 years.

Drug interactions

  • antihypertensive agents (especially those that affect the sympathetic nervous system): various cardiovascular effects may develop;
  • tricyclic antidepressants: the risk of arrhythmias increases;
  • vasoconstrictor drugs, some antidepressants (MAO inhibitors, tricyclic antidepressants): blood pressure may rise.
