Release form and composition
The product is produced in the form of tablets: flat-cylindrical, yellow with a green tint, with a chamfer (6, 10 or 12 pieces in a blister strip packaging; 50 pieces in a glass or polymer jar; in a cardboard box instructions for using Doverin and 1, 2, 3, 4 or 5 packs or 1 can).
1 tablet contains:
- active substance: drotaverine hydrochloride - 40 mg;
- additional components: potato starch, lactose monohydrate (milk sugar), magnesium stearate, water-soluble methylcellulose, talc.
Pharmacological properties
Pharmacodynamics
Drotaverine is a myotropic antispasmodic derived from isoquinoline. The active substance inhibits the enzyme type IV phosphodiesterase (PDE-4), resulting in the accumulation of intracellular cyclic adenosine monophosphate (cAMP) and, as a result, inactivation of myosin light chain kinase, which provides relaxation of smooth muscle tissue.
The use of the drug is effective for smooth muscle spasms of neurogenic and muscular etiology. Trust, regardless of the type of autonomic innervation, has an effect on the smooth muscles of the genitourinary, biliary and vascular systems, as well as the gastrointestinal tract (GIT). In the myocardium and vessels of smooth muscles, the cAMP hydrolyzing enzyme is mainly PDE-3, and therefore, despite the high antispasmodic activity of drotaverine hydrochloride, its significant therapeutic effect on the cardiovascular system (CVS) and the occurrence of serious adverse reactions from the latter are not recorded. …
Pharmacokinetics
Doverin is intensively absorbed in the digestive tract. After oral administration, the half-absorption period of drotaverine is 12 minutes, the bioavailability is almost 100%. The agent is characterized by a uniform distribution over the tissues, it penetrates into the cells of smooth muscles, but does not pass through the blood-brain barrier. The active substance is mostly excreted in the urine and to a small extent in the bile.
Indications for use
Doverin is indicated for the relief of smooth muscle spasms caused by the following diseases:
- cholecystitis, pericholecystitis, cholecystolithiasis, cholangiolithiasis, papillitis and other lesions of the biliary tract;
- ureterolithiasis, nephrolithiasis, pyelitis, tenesmus of the bladder, cystitis and other pathologies of the urinary tract.
As a means of auxiliary therapy, the drug is recommended for use in the following cases:
- tensor headache;
- spasms of smooth muscles of the gastrointestinal tract in diseases such as gastritis, peptic ulcer and 12 duodenal ulcer, enteritis, spasms of the cardia and pylorus, colitis, including mucous colitis with flatulence and spastic colitis, accompanied by constipation;
- gynecological diseases, including dysmenorrhea.
Contraindications
Absolute:
- severe renal and / or hepatic impairment;
- low cardiac output syndrome (severe heart failure);
- age up to 18 years;
- lactation period;
- glucose-galactose malabsorption, lactase deficiency, lactose intolerance;
- hypersensitivity to any component of the antispasmodic agent.
Relative (you must use Doverin tablets with extreme caution):
- severe atherosclerosis of the coronary arteries;
- arterial hypotension;
- glaucoma;
- hyperplasia of the prostate;
- period of pregnancy.
Doverin, instructions for use: method and dosage
Doverin tablets are taken orally.
Recommended dosage regimen: 1-2 tablets (40-80 mg) 2-3 times a day.
Side effects
- CVS: lowering blood pressure, heart palpitations;
- nervous system: dizziness, insomnia, headache;
- Gastrointestinal tract: nausea, constipation;
- immune system: allergic reactions, including allergic dermatosis.
Overdose
Symptoms of an overdose of drotaverine can be atrioventricular block, paralysis of the respiratory center, cardiac arrest.
In the case of oral use of an excessive dose of the drug, it is necessary to stop taking Doverin and conduct symptomatic therapy. There is no specific antidote.

In the treatment of gastric ulcer and duodenal ulcer, the antispasmodic agent should be used in combination with antiulcer drugs.
Influence on the ability to drive vehicles and complex mechanisms
Drotaverine, taken orally in therapeutic doses, does not affect the ability to drive and perform work requiring increased attention. If any undesirable reactions develop, the question of driving vehicles and working with mechanisms is subject to individual consideration. If dizziness occurs after taking the pills, you should avoid engaging in potentially hazardous activities, including driving and operating machinery.
Application during pregnancy and lactation

Pediatric use

Drug interactions
- other antispasmodics (including M-anticholinergics): the effect of these funds is enhanced;
- quinidine, procainamide, tricyclic antidepressants: the risk of arterial hypotension associated with the use of these drugs is aggravated;
- morphine: its spasmogenic activity decreases;
- phenobarbital: the spasmolytic effect of drotaverine increases;
- levodopa: the antiparkinsonian effect of this substance is weakened.
