Release form and composition
Dazolik is produced in the form of film-coated tablets: almost white or white, biconvex, round, with a line on one side; at the fracture - the core is almost white or white, surrounded by a shell (10 pcs. in blisters, 1 or 5 blisters in a cardboard box).
The composition of 1 tablet includes:
- Active ingredient: ornidazole - 500 mg;
- Auxiliary components: starch, purified talc, microcrystalline cellulose, sodium carboxymethyl starch (type A), sodium lauryl sulfate, titanium dioxide, povidone K90, magnesium stearate, colloidal silicon dioxide, copolymer of butyl methacrylate, methyl methacrylate 1: 1 dimethacylamino [E100]), macrogol 6000.

Ornidazole is a derivative of 5-nitroimidazole, the mechanism of action of which is the biochemical reduction of the 5-nitro group of ornidazole, carried out by intracellular transport proteins of protozoa and anaerobic bacteria. The restored 5-nitro group of the active component of Dazolik acts on the DNA of cells of pathogens, inhibiting the production of their nucleic acids, which contributes to the destruction of bacteria.
Ornidazole is active against Clostridium spp., Trichomonas vaginalis, Fusobacterium spp., Entamoeba histolytica, Giardia lamblia, as well as against the anaerobic bacteria Bacteroides spp. (including Bacteroides vulgates, Bacteroides fragilis, Bacteroides thetaiotaomicron, Bacteroides ovatus, Bacteroides distasonis). Aerobic microorganisms show resistance to ornidazole.
Pharmacokinetics
Ornidazole is highly absorbed from the gastrointestinal tract when taken orally. Its bioavailability is 90%. The maximum concentration of the substance in the blood is reached within 1–2 hours. Ornidazole binds to plasma proteins by less than 15% and is metabolized in the liver through glucuronidation, oxidation and hydroxylation. The half-life is 12-14 hours.
Ornidazole crosses the placental and blood-brain barriers, and is also determined in breast milk and most tissues. It is excreted in the form of metabolites in the urine (approximately 60–70% of the dose taken) and with feces (about 20–25% of the dose taken). Approximately 5% of the dose of ornidazole is excreted unchanged.
Indications for use
- Trichomoniasis;
- Giardiasis;
- Amoebic dysentery, amebiasis, extraintestinal amebiasis (including amoebic liver abscess);
- Infections caused by anaerobic bacteria (for prophylaxis during operations in gynecology and on the colon).
Contraindications
- Age up to 3 years;
- Hypersensitivity to drug components.
According to the instructions, Dazolik must be taken with caution in lactating and pregnant women, as well as in diseases of the central nervous system (including epilepsy, multiple sclerosis), alcoholism and liver diseases.
Instructions for use of Dazolik: method and dosage
Dazolik tablets should be taken orally after meals.
The scheme of drug use is determined by the indications:
- Trichomoniasis: adults - a single dose of 1500 mg or 1000 mg in combination with intravaginal administration of 500 mg vaginal tablets at night. You can carry out therapy for 5 days, taking the drug orally 2 times a day (in the morning and in the evening), 500 mg simultaneously with the use of 1 vaginal tablet of 500 mg at night. For children, Dazolik is prescribed once, setting the dose at the rate of 25 mg / kg of body weight;
- Giardiasis: adults and children weighing 35 kg - 1500 mg once a day, children from 3 years old and weighing up to 35 kg - 40 mg / kg per day. The duration of therapy is 1-2 days;
- Amebic dysentery: children weighing 35 kg and adults - 1 time a day, 1500 mg, with a body weight of 60 kg - 2000 mg per day. For children from 3 years old and weighing up to 35 kg, the daily dose is determined at the rate of 40 mg / kg of body weight. The duration of therapy is 3 days;
- Other forms of amebiasis: children from 12 years old and adults - 500 mg 2 times a day; children from 3 years old and weighing up to 35 kg - at the rate of 25 mg / kg of body weight, frequency of application - 1 time per day. The duration of therapy is 5-10 days.
For the prevention of infections caused by anaerobic bacteria, Dazolic is prescribed 500-1000 mg before the operation and 2 times a day, 500 mg for 3-5 days after the operation.
Side effects
During therapy, the following side effects may develop:
- Digestive system: increased activity of liver enzymes, nausea;
- Central and peripheral nervous system: temporary loss of consciousness, headache, drowsiness, peripheral neuropathy (mixed or sensory), tremor, dizziness, muscle stiffness, convulsions, impaired coordination of movements, fatigue, perversion of taste sensations;
- Others: allergic reactions.
Overdose
Signs of an overdose of Dazolik include peripheral neuritis, depression and seizures. In this case, symptomatic therapy is recommended, and in case of convulsions, diazepam may be required.

With trichomoniasis, it is necessary to carry out simultaneous therapy for both sexual partners.
Since ornidazole has a depressing effect on the central nervous system, it should not be used while driving or when performing work that requires precise coordination of movements.
Application during pregnancy and lactation
Dazolik tablets are not recommended for use during lactation and pregnancy, especially in the first trimester.
Drug interactions
Unlike other imidazole derivatives (metronidazole), Dazolik does not inhibit acetaldehyde dehydrogenase and is therefore compatible with ethanol.
With the simultaneous use of ornidazole can enhance the effect of indirect anticoagulants of the coumarin series and lengthen the muscle relaxant effect of vecuronium bromide.
