Alpha-tocopherol (vitamin E) is the most effective of tocopherols - biologically active substances that protect the intermediate compounds of cellular respiration from oxidation of the functional groups of their molecules. They have the ability to oxidize to semiquinones, which are reduced to tocopherols in the body. If their amount in the human body is sufficient, they protect the metabolites of cellular respiration and unstable mediators from oxidation. They play a buffer role in redox processes at the level of cellular metabolism under pathological conditions.

Vitamin E, entering the body, binds free oxygen radicals, acts as a cellular antioxidant, prevents erythrocyte hemolysis, participates in cell proliferation, protein and heme biosynthesis, as well as other important processes of tissue metabolism. Alpha-tocopherol strengthens the walls of blood vessels, is involved in the development and functioning of connective tissue.

Being a natural antioxidant, vitamin E inhibits lipid peroxidation by free radicals, is used to maintain normal erythrocyte resistance, and activates phagocytosis. In large doses, this vitamin is able to prevent platelet aggregation.

Pharmacokinetics

Alpha-tocopherol is absorbed mainly from the duodenum - from 20 to 40%, important conditions for its absorption are the normal functioning of the pancreas, the presence of fats and bile salts. The degree of absorption decreases with increasing dose of the drug.

The maximum concentration reaches within 4 hours. It accumulates in all tissues (especially in adipose tissue) and organs.

Vitamin E crosses the placenta in small quantities: 20-30% of the concentration in the mother's blood enters the fetal blood. Penetrates into breast milk.

It is excreted mainly through the gastrointestinal tract with bile - over 90%. Less than 6% of the substance is excreted by the kidneys in the form of metabolites, including glucuronides.

Indications for use

Vitamin E Zentiva is used to prevent and treat vitamin E deficiency, including in the following cases:

  • heavy physical activity;
  • the recovery period after serious illnesses, operations, injuries;
  • menstrual irregularities (as part of complex hormonal therapy);
  • chronic hepatitis (as part of combination therapy);
  • proliferative and degenerative changes in the musculoskeletal system: amyotrophic lateral sclerosis, muscular dystrophy (as part of complex therapy).

Contraindications

  • acute myocardial infarction;
  • childhood;
  • increased sensitivity to the components of Vitamin E Zentiva.

The drug should be used with caution in the presence of an increased risk of thromboembolism, after myocardial infarction, in severe cardiosclerosis.

At a dose of more than 400 IU (1.21 IU - 1 mg), vitamin E can worsen the course of hypothrombinemia caused by vitamin K deficiency, so caution is also required in this case.

Instructions for the use of Vitamin E Zentiva: method and dosage

The drug is taken orally.

Vitamin E Zentiva is prescribed only for adults.

Recommended dosage regimens:

  • hypovitaminosis E: 100 mg per day until the condition normalizes;
  • increased physical activity: 100 mg per day for the entire period of stress;
  • degenerative and proliferative changes in the musculoskeletal system: 100 mg per day for 1–2 months;
  • chronic hepatitis: 100 mg once a day;
  • recovery period after serious illness / surgery / injury: 100 mg per day for 1-2 weeks;
  • menstrual irregularities: 300-400 mg every other day in succession, starting from the 17th day of the cycle until the last day of the cycle (the day before the first day of menstruation).

Side effects

Vitamin E Zentiva, taken at the recommended dosage, usually does not cause side effects.

In some cases, it is possible:

  • from the gastrointestinal tract: gastralgia, nausea, diarrhea;
  • from the vascular system: pulmonary embolism, thrombophlebitis;
  • on the part of metabolism and nutrition: an increase in the concentration of cholesterol in the blood serum, an increase in the activity of creatine phosphokinase, hypercreatinuria;
  • from the immune system: allergic reactions.

Overdose

With prolonged intake of high doses of vitamin E (400-800 mg per day), the following symptoms of overdose may occur: visual impairment, dizziness, headache, fatigue, fainting, diarrhea, nausea, with epidermolysis bullosa - the growth of white hair in areas of alopecia.

With prolonged use of very high doses of alpha-tocopherol (more than 800 mg per day) in patients with vitamin K deficiency, bleeding is possible. In sensitive patients, there is a risk of impaired metabolism of thyroid hormones, the likelihood of developing thromboembolism and thrombophlebitis increases.

If symptoms of an overdose appear, Vitamin E Zentiva is canceled, glucocorticosteroids are prescribed and symptomatic therapy is performed.

In the case of taking vitamin E in high daily doses (more than 400 IU), the deficiency in the body of vitamin K may increase, which causes hypoprothrombinemia. In addition, the effect of simultaneously used anticoagulants is enhanced. In this regard, in order to avoid an overdose, you should not take any other drugs in combination, which contain alpha-tocopherol.

Patients with congenital epidermolysis bullosa are likely to grow white hair in areas affected by alopecia.

Vitamin E enhances the action of cardiac glycosides, non-steroidal anti-inflammatory drugs, glucocorticosteroids.

The absorption of vitamin E is reduced with the simultaneous use of colestipol, cholestyramine and mineral oils.

In patients receiving iron supplements in high doses, oxidative processes in the body increase, therefore, the need for vitamin E increases.

To avoid overdose, it is not recommended to take other vitamin complexes containing alpha-tocopherol simultaneously with Vitamin E Zentiva.
