Vegetrox is a drug with antihypertensive and antipruritic action.

Release form and composition

The drug is produced in the form of tablets: biconvex round, white with a yellow tint or white (10 pcs. In a blister strip of aluminum foil and polyvinyl chloride film; 10, 20 or 30 pcs. In a polymeric polyethylene test tube, sealed with a stopper; 30, 40, 50 or 100 pcs. In a polymeric polyethylene jar with a lid equipped with a compensator; in a cardboard box 1, 2, 3, 4 or 5 blisters or 1 test tube / jar and instructions for the use of Vegetrox).

1 tablet contains:

  • active substance: proxane hydrochloride - 0.015 g;
  • additional components: magnesium stearate, potato starch, talc, lactose monohydrate.

Pharmacological properties

Pharmacodynamics

Prooxan has antihypertensive and antipruritic effects. The active substance of Vegetrox blocks postsynaptic α 1 - and presynaptic α 2-adrenergic receptors, regardless of their location. Demonstrates peripheral and central influence, reduces the sensitivity of adrenergic receptors to adrenaline, leading to a change in the permeability of cell membranes. Reduces the intensity of stimulation of the hypothalamic structures, reducing the production of tropic hormones and hormones of the peripheral glands. Proproxan helps to weaken the transmission of adrenergic vasoconstrictor impulses, eliminate spasm and expand peripheral vessels, especially arterioles and precapillaries. Improves blood supply to mucous membranes, skin and muscles; inhibits the excitability of the diencephalic structures of the brain and participates in the regulation of the tone of the sympathoadrenal system.

Pharmacokinetics

When taken orally, propoxan is quite well absorbed from the digestive tract, the effective concentration is noted after 30–40 minutes and lasts for 3–4 hours. The active substance is characterized by a weak bond with plasma proteins and a large volume of distribution (V d), intensively distributed in various tissues and body fluids, including cerebrospinal fluid.

Proproxan overcomes blood-brain and other histohematogenous barriers well; selectively retained in the hypothalamus, in which the posterior region contains mainly noradrenergic neurons. The drug is excreted mainly by the kidneys, approximately 70% of the administered dose - in the first 24 hours.

In preclinical experiments with intraperitoneal administration in animals, the half-life (T 1/2) of propoxan was not established. At the same time, 1 hour after the injection of the drug, less than 5% of the administered dose was found in the blood plasma, and after 2 hours only traces were found. After 1 hour, 22% of the maximum concentration (C max) of the agent was detected in the brain, and after 2 hours - 6%. In the course of experiments with carbon-labeled propoxan, it was also found that the products of its metabolism can be retained for up to 10 days in various tissues and organs (plasma, liver, kidneys, brain).

Indications for use

  • somatoform autonomic dysfunction of the sympathoadrenal type, including proceeding with an increase in blood pressure (BP) (as a symptomatic agent in combination therapy);
  • allergic dermatitis (as a symptomatic agent in combination therapy for relieving itching).

Contraindications

Absolute:

  • circulatory failure II – III degree;
  • ischemic heart disease (IHD) of the II – IV functional class;
  • cardiogenic shock;
  • myocardial infarction;
  • bradycardia;
  • severe atherosclerosis;
  • arterial hypotension;
  • severe cerebrovascular insufficiency (including a history of indications of strokes, transient ischemic attacks);
  • syndrome of glucose-galactose malabsorption, lactose intolerance, lactase deficiency;
  • age up to 18 years;
  • hypersensitivity to any ingredient of the drug.

Relative (you should take Vegetrox tablets with extreme caution): stomach ulcer, coronary artery disease, arrhythmia, old age.

During the period of taking Vegetrox, blood glucose levels should be monitored, in view of a possible increase in the content of endogenous insulin.

Vegetrox, instructions for use: method and dosage

Vegetrox tablets are taken orally. A single dose of the drug is 15-30 mg of propoxan (1-2 tablets).

Recommended frequency of administration in combination therapy as a symptomatic drug:

  • somatoform autonomic dysfunction, proceeding according to the sympathoadrenal type: 2-3 times a day;
  • allergic dermatitis (to relieve itching): 1 time per day before bedtime.

The duration of treatment with Vegetrox is established by the attending physician.

Side effects

  • gastrointestinal tract: with an unknown frequency - an increase in the production of hydrochloric acid in the stomach, diarrhea;
  • cardiovascular system: with an unknown frequency - nasal congestion, hyperemia of the skin, dizziness, arterial hypotension, bradycardia, arrhythmias, orthostatic hypotension, the occurrence of angina attacks in the presence of a predisposition;
  • skin: with an unknown frequency - allergic reactions.

Overdose

Symptoms of a proxan overdose may include reactions such as a sharp drop in blood pressure, bradycardia, orthostatic collapse.

In this condition, symptomatic treatment is prescribed, aimed at restoring and maintaining vital functions.

After taking the first dose of the drug, the following reactions are possible: weakness, headache, palpitations, fainting, a sharp decrease in blood pressure, worsening of the subjective state (the phenomenon of the first dose), which does not require the abolition of Vegetrox, since this effect disappears with repeated administration of the tablets. Drowsiness and a short-term feeling of numbness of the tongue are also possible.

In addition, the first dose requires blood glucose control due to the risk of increased endogenous insulin levels. As a result of the above, treatment should be started with the minimum effective dose, under the supervision of a physician, and for 6-8 hours after taking Vegetrox, observe bed rest.

Influence on the ability to drive vehicles and complex mechanisms

During the period of therapy with Vegetrox, it is not recommended to drive a car and engage in other activities that are potentially dangerous and require increased concentration of attention and increased speed of reactions.

Application during pregnancy and lactation

The use of Vegetrox during pregnancy and lactation is contraindicated due to insufficient data on its effectiveness and safety in pregnant and lactating women.

Pediatric use

Prescribing the drug to patients under 18 years of age is contraindicated due to the lack of data confirming the effectiveness and safety of using Vegetrox in persons belonging to this age group.

Use in the elderly

Elderly patients should be treated with caution with Vegetrox.

Drug interactions

  • antihypertensive drugs: decrease in blood pressure is potentiated;
  • adrenaline: a weakening of the effect of this substance is possible (the appearance of a depressor effect instead of a pressor effect);
  • antipsychotics (derivatives of butyrophenone, diphenylbutylpiperidine, phenothiazine, thioxanthene): the effect of these agents is enhanced;
  • novocainamide: there may be a decrease in the value of the stroke volume of the heart.
