Pharmacological properties

Pharmacodynamics

Memantine is an adamantane derivative and a noncompetitive antagonist of N-methyl-D-aspartate (NDMA) receptors. It is characterized by a modulating effect on the glutamatergic system. It is responsible for the regulation of ion transport, is a blocker of calcium channels, normalizes membrane potential, activates the processes of transmission of nerve impulses. Akatinol Memantine has a positive effect on cognitive processes and enhances daily activity.

Pharmacokinetics

When taken orally, memantine is completely absorbed at a fairly high rate. Its maximum level in blood plasma is reached within 3-8 hours. With normal renal function, the accumulation of the substance in the body is not observed.

When Akatinol Memantine is taken in a daily dose of 20 mg once a day, the equilibrium plasma concentration of memantine is 70–150 ng / ml. Approximately 80% of the active ingredient of the drug is excreted unchanged. In vitro studies have shown that the metabolism, carried out with the participation of the cytochrome P450 system, is not observed. Memantine metabolites are not characterized by pharmacological activity.

The compound is excreted from the body monoexponentially, and the half-life in the terminal phase is 60–100 hours. Memantine is excreted through the kidneys. When the pH of urine is above 7, the rate of its excretion decreases.

Reviews about Akatinol Memantine

There are various reviews about Akatinol Memantine: both positive and negative. For elderly patients diagnosed with Alzheimer's disease or another variant of dementia, the drug is considered indispensable. However, it should be understood that he is only able to slow down the development of the disease and slightly improve the general well-being of the patient. To date, drugs that completely relieve a person from such diseases have not yet been found, and Akatinol Memantine is no exception. Many patients also dislike the very high price of the drug.

Sometimes a drug is prescribed to children in order to improve their mental development, despite the fact that the drug is contraindicated for this category of patients in accordance with the instructions. Reviews about him when used for these purposes range from enthusiastic to sharply negative. Many parents argue that the initially positive influence on the child is often replaced by a sharp regression, which returns the little patient to his original state or even worsens it. Therefore, the decision to use Akatinol Memantine in children should be made after consultation with a specialist and with the full understanding of the parents of the possible consequences.
