Release form and composition
The drug is produced in the form of eye drops: colorless or slightly colored transparent liquid (0.4 ml each in a dropper tube made of polyethylene or polypropylene, in a foil bag 5 or 10 pcs., In a cardboard box 2, 4, 6 bags of 5 pcs. or 1, 2, 3 bags of 10 pcs. and instructions for the use of SIGIDA duo).
1 ml of the preparation contains:
- active ingredients: naphazoline nitrate (naphthyzine) - 0.33 mg; diphenhydramine hydrochloride - 1 mg;
- additional components: 0.1 M nitric acid solution or 1 M sodium hydroxide solution, macrogol 300, disodium edetate dihydrate (Trilon B), boric acid, sodium hyaluronate, water for injection.

Pharmacodynamics
SIGIDA duo is a combined drug for topical use in ophthalmology, which has antihistamine and vasoconstrictor effects.
Diphenhydramine is a histamine H 1 receptor antagonist. Through a competitive blockade of the latter, the active substance reduces the severity of allergic symptoms, especially those caused by the release of histamine, which include increased permeability and vasodilation. Reduces itching, swelling, watery eyes.
Naphazoline is an α-adrenergic agonist that stimulates vascular α-adrenergic receptors. When used topically, the active ingredient leads to a prolonged narrowing of peripheral vessels, reduces the symptoms of an inflammatory condition, and has an anti-edema effect.
Pharmacokinetics
In full, the local effect of naphazoline begins to appear after 5 minutes from the moment of use and is observed within 6-8 hours. unlikely. The risk of systemic reactions is highest in young children and elderly patients.
When applied topically, the likelihood of a systemic effect of diphenhydramine is extremely low.
Indications for use
- seasonal or perennial allergic conjunctivitis in the acute phase;
- inflammatory conditions caused by irritation of the conjunctiva due to wearing contact lenses, exposure to cigarette smoke, sunlight, contact with water in a swimming pool, etc.
Contraindications
Absolute:
- angle-closure glaucoma;
- age up to 2 years;
- pregnancy and the period of breastfeeding;
- hypersensitivity to any component of the antiallergic agent (including sympathomimetic amines).
Relative (use SIGIDA duo eye drops with great care):
- prolonged conjunctivitis (drops can be used only for a short period against the background of an exacerbation of a chronic disease);
- elderly age.
If symptoms of irritation or pain in the eyes persist against the background of therapy for more than 72 hours, the drug should be discontinued.
Despite the fact that when instilling SIGIDA duo into the conjunctival sac, the systemic effect of its components is unlikely, it is necessary to use drops with caution if patients have the following diseases: atherosclerosis, hypertension, arrhythmia, chronic rhinitis, bronchial asthma, hyperthyroidism, prostatic hyperplasia. Such patients urgently need to stop using drops if any systemic reaction develops, indicating the absorption of naphazoline.
It is not recommended to carry out drug treatment for children aged 2 to 6 years.
SIGIDA duo, instructions for use: method and dosage
Eye drops SIGIDA duo are applied topically as instillation into the conjunctival sac.
Children over 6 years of age, adolescents and adults are recommended to inject the drug, if necessary, every 6-8 hours, 1-2 drops. Do not use the product longer than 3-5 days.
The dose of the drug contained in 1 dropper tube is sufficient for one instillation in both eyes.
The order of administration of drops:
- Open the bag and separate 1 dropper tube, put the rest back into the bag.
- After making sure that the droplets are at the bottom of the dropper tube, open it by rotating and separating the valve with rotating movements.
- Inject the required dose of the drug into the eyes.
- Throw away the dropper tube after instillation, even if there is a solution in it.
Side effects
During the period of application of SIGIDA duo, the following adverse reactions may occur: dryness of the nasal mucosa, hyperemia, burning, itching, irritation of the conjunctiva, visual disturbances, eye pain, mydriasis, increased intraocular pressure. Against the background of treatment with the agent for 7 days and with a frequency of administration of at least 10 times a day, 1 case of corneal opacity was recorded. This effect was removed after discontinuation of the drug.
A long course of therapy can lead to local changes in the epithelium caused by hypoxia (worsening prognosis).
Overdose
Too frequent or prolonged use of SIGIDA duo in children under 6 years of age can cause the appearance of disorders such as hypothermia, prolonged mydriasis, inhibition in the central nervous system (CNS), increased blood pressure (BP), tachycardia, coma. Treatment is symptomatic.

The use of an antiallergic agent for more than 5 days or with an interval of less than 3 hours is contraindicated due to the threat of the development of a syndrome that causes a secondary increase in edema and hypersecretion, as well as the occurrence of persistent changes in the epithelium.
Drops are intended exclusively for topical use - introduction into the conjunctival sac.
Influence on the ability to drive vehicles and complex mechanisms
Patients driving vehicles or servicing other complex mechanisms should use SIGIDA duo with extreme caution because of the possible appearance of visual impairment.
Application during pregnancy and lactation
The use of SIGIDA duo is contraindicated for pregnant women.
If necessary, course administration of drops during lactation, it is required to stop breastfeeding.
Pediatric use
It is contraindicated for children under 2 years old to use SIGIDA duo, for children aged 2 to 6 years old it is not recommended due to the possible occurrence of potentially dangerous adverse events.
Use in the elderly
Elderly patients should use SIGIDA duo with caution due to the increased risk of systemic exposure to its active substances.
Drug interactions
- tricyclic antidepressants: the vasoconstrictor effect of naphazoline is enhanced;
- monoamine oxidase (MAO) inhibitors: the risk of developing a hypertensive crisis is aggravated when naphazoline is combined with these drugs, as well as when it is used within 10 days after the end of their intake.
