Release form and composition

The dosage form of Selanka is nasal drops 0.15%: transparent, colorless (3 ml each in glass bottles with a pipette stopper or bottles equipped with a pipette with a lid, in a cardboard box 1 bottle or 1 set).

Composition of 1 ml drops:

  • active substance: threonyl-lysyl-prolyl-arginyl-prolyl-glycyl-proline diacetate (in terms of 100% substance) - 0.001 5 g;
  • auxiliary substances: purified water - up to 1 ml; nipagin (methyl parahydroxybenzoate) - 0.001 g.

Pharmacological properties

Pharmacodynamics

Selank is a synthetic analogue of taftcin (endogenous peptide), has an original mechanism of neurospecific action on the central nervous system (CNS), binds to specific receptors on the membranes of nerve cells.

The drug affects the activity of the brain enzymes tyrosine and tryptophan hydroxylase, the exchange of monoamines in the emotiogenic structures of the brain (cerebral cortex, diencephalon, hypothalamus).

By normalizing the level of serotonin in the brain under conditions of an experimentally induced decrease, it exhibits tropism to the serotonergic system.

Increases the resistance of cerebral cortex neurons to high-intensity functional loads, stabilizes the processes of inhibition and excitation in the brain.

In the spectrum of its pharmacological action, anxiolytic effect with an activating (stimulating) component prevails. It does not have a hypnosedative and muscle relaxant effect. It has a positive effect on the cognitive and mnestic functions of the brain, including when they are impaired. Improves the parameters of attention and short-term memory, activates the processes of reproduction and analysis of information, memory and learning.

Increases motivational stability and the adequacy of adaptive behavior. It has a vegetative effect, improves the vegetative support of activity during emotional stress, and has an optimizing effect on the body's adaptive reserve.

In comparison with ED50, the drug does not reveal any undesirable side and toxic effects at 200-300 fold increase in the dose. Selank has no undesirable long-term effects, does not have a local irritating, allergenic, teratogenic and embryotoxic effect, does not have mutagenic properties, does not cause drug dependence.

Pharmacokinetics

With intranasal administration, the absolute bioavailability of the drug is 92.8%. It is rapidly absorbed from the nasal mucosa into the bloodstream and is found in it half a minute after injection. A progressive decrease in its concentration in blood plasma is observed for 5–5.5 minutes.

With intranasal administration, drug metabolites are not detected. Selank penetrates into the brain tissue, quickly spreads to tissues and organs, and is found in well-vascularized organs (such as the heart, kidneys and liver) unchanged.

Due to the rapid degradation under the influence of tissue peptidases, neither metabolites nor unchanged drug is detected in daily urine.

Indications for use

According to the instructions, Selank is recommended for the treatment of the following anxiety conditions:

  • panic attacks;
  • emotional instability;
  • sleep disorders;

Instructions for the use of Selank: method and dosage

Drops are applied intranasally, in a sitting position with the head slightly thrown back. After instillation, each nostril is clamped with a finger for a short time.

Before use, the tip of the pipette on the dropper bottle is carefully cut along the cut line. Then a cap is put on the pipette (it must close it tightly). The bottle is turned over and lightly tapped on the bottom with a finger so that the liquid moves to the pipette and fills its entire space. After that, the cap is removed and, by slightly pressing on the wide part of the pipette, the required number of drops are squeezed into the nasal passage.

With increased secretion of the secretion of the mucous membrane of the nasal cavity, the effectiveness of absorption of the drug may decrease, and therefore, the nasal passages should be cleaned before instillation.

Recommended dosing regimen: 2 drops of Selanka three times a day in each nasal passage. The duration of the course is 2 weeks. If necessary, in agreement with the attending physician, the treatment can be repeated, but not earlier than after 1–3 weeks.

Side effects
