Trizistone is a combined (estrogen-gestagenic) three-phase oral contraceptive.

Release form and composition

Triziston is produced in the form of pills: three types in one package - red-brown, white and mustard color (21 pcs. (6 pcs. Red-brown and white and 9 pcs. Mustard color) in a blister strip, in a cardboard pack 1 or 3 packs).

The active ingredients are ethinyl estradiol and levonorgestrel, their content is in 1 tablet (respectively): red-brown color - 0.03 mg and 0.05 mg, white color - 0.04 mg and 0.075 mg, mustard color - 0.03 mg and 0.125 mg.

Pharmacological properties

Pharmacodynamics

Trizistone is a combined estrogen-progestin oral contraceptive. The estrogenic component is ethinyl estradiol, the gestagenic component is levonorgestrel (a derivative of 19-nortestosterone). The therapeutic effect of the drug is associated with the inhibition of the pituitary secretion of gonadotropic hormones.

In terms of activity, levonorgestrel is superior to the corpus luteum hormone progesterone and its synthetic analogue pregnin. The mechanism of its action is carried out without preliminary metabolic transformations at the receptor level. The influence of levonorgestrel provides a blockade of the release of releasing factors - follicle-stimulating and luteinizing hormones of the hypothalamus. As a result of inhibition of the secretion of gonadotropic hormones by the pituitary gland, the maturation of the egg and its release from the ovary slows down. Ethinylestradiol enhances the contraceptive effect of the drug, maintaining a high viscosity of cervical mucus. This protects the uterine cavity from free entry of sperm.

Regular use of Trizistone, in addition to the contraceptive effect, helps to normalize the menstrual cycle, prevent the development of neoplasms and other gynecological diseases.

Pharmacokinetics

After oral administration, the active substances are absorbed from the gastrointestinal tract quickly and completely. The maximum concentration of ethinyl estradiol in blood plasma is reached after 1.5 hours, levonorgestrel - after 2 hours.

The metabolism of active substances occurs in the liver.

The half-life of the drug is from 2 to 7 hours.

Ethinyl estradiol and levonorgestrel are excreted in breast milk.

60% of levonorgestrel and 40% of ethinyl estradiol are excreted through the kidneys, 40% and 60%, respectively, through the intestines.

Indications for use

Monophasic dosage forms are used to treat hormone-dependent functional disorders of the menstrual cycle, including premenstrual syndrome, dysmenorrhea, menorrhagia without an organic cause.

Contraindications

  • congenital hyperbilirubinemia (Gilbert's syndrome, Dubin-Johnson syndrome, Rotor syndrome);

Instructions for the use of Triziston: method and dosage

Dragee Triziston is taken orally, swallowed whole and washed down with a sufficient amount of liquid.

With contraception, the woman determines the time of admission for herself independently and strictly observes it throughout the course. It is preferable to take the drug after breakfast or dinner. Since the contraceptive effect is ensured by maintaining a constant concentration of hormones in the blood plasma, the interval between doses should be from 22 to 26 hours and not exceed 36 hours.

Dragee take 1 pc. 1 time per day, starting from the 1st or 5th day of menstrual bleeding [depending on the type of drug used: monophasic (in a package of dragees of the same color), two- or three-phase (in a package of dragees of different colors)], daily. Contraception should be started by taking red-brown pills, then white pills are taken for 6 days and mustard pills for 9 days. After 21 days, when all the pills from the package are taken, there is a break in taking the drug for 7 days, during which menstrual bleeding passes. After a 7-day break, even if the bleeding continues, you should start taking the red-brown dragee from a new package.

If you accidentally skip taking the next dose of the drug at the set time, the pills should be taken as soon as you remember. If the delay does not exceed 12 hours, then the contraceptive effect of Triziston remains. If the missed dose is taken in a period exceeding 36 hours from the moment of taking the previous tablet, the contraceptive effect is impaired. In this regard, until the end of the menstrual cycle, taking pills should be continued and at the same time use barrier methods of contraception.

If, after taking the next dose, a woman has vomiting or diarrhea, there is a risk of incomplete absorption of the drug into the blood and hormonal imbalance. If the symptoms of discomfort disappear within one day, it is recommended to take an additional 1 tablet from the package and continue the cycle. If vomiting and diarrhea continue for several days, then the contraceptive effect of the drug is impaired and barrier contraceptive methods should be used.

When prescribing Triziston for therapeutic purposes, the doctor determines the dose individually. Usually it is 2-3 pcs. in a day. The duration of the course of therapy is 10 days or until the bleeding stops completely.

Side effects
