Release form and composition

The drug is produced in the form of film-coated tablets: biconvex, round; the film shell is orange, the core in the cross section is orange with small white blotches (6 or 10 pieces in a blister, 20 pieces in a polymer can / bottle; in a cardboard box 1 blister or 1 can / bottle and instructions for use of Tilorona-SZ).

1 film-coated tablet contains:

  • active substance: tilorone (in the form of dihydrochloride) - 125 mg;
  • additional components (core): hypromellose (hydroxypropyl methylcellulose), microcrystalline cellulose, hyprolose (hydroxypropyl cellulose), potato starch, magnesium stearate;

Pharmacological properties

Tilorone is a low molecular weight synthetic interferon inducer (IFN) that stimulates the body to produce all types of IFNs - α, β, γ, λ. As a result of the action of the active substance, hepatocytes, intestinal epithelial cells, T-lymphocytes, granulocytes and neutrophils become the main producers of IFN. After oral administration, the peak of IFN formation is recorded 4–24 hours later in the intestine-liver-blood sequence.

Tilorone exhibits immunomodulatory and antiviral effects. After a single oral administration of the active substance at a dose equal to the maximum human dose per day, the highest content of IFN-λ in the lung tissue was detected after 24 hours, and IFN-α - after 48 hours. Induction of IFN-λ in the lung tissue contributes to the enhancement of antiviral protection the respiratory tract for influenza and other respiratory infections caused by viruses.

In human leukocytes, tilorone induces IFN production. It stimulates stem cells in the bone marrow, in accordance with the dose, increases antibody production, reduces the degree of immunosuppression, restores the ratio of two subtypes of T cells - T-suppressors and T-helpers. Tiloron-SZ demonstrates effectiveness against various viral infections, including acute respiratory viral infections (ARVI), including influenza, as well as herpesvirus infections and viral hepatitis. The mechanism of antiviral action is based on the suppression of translation of virus-specific proteins in infected cells, which leads to inhibition of viral reproduction.

Pharmacokinetics

After oral administration, the agent is rapidly absorbed from the gastrointestinal tract (GIT), bioavailability is 60%. The drug binds to plasma proteins by about 80%. Almost unchanged, it is excreted through the intestines and kidneys - 70 and 9%, respectively. The half-life (T 1/2) is 48 hours, the agent does not undergo biotransformation and does not accumulate in the body.

Tiloron-SZ tablets should be taken orally after meals.

Recommended dosage regimen:

  • influenza and other acute respiratory viral infections: for medicinal purposes - the first 2 days of therapy at 125 mg / day, then after 48 hours at 125 mg, course dose - 750 mg (6 tablets); for prophylaxis - 125 mg once a week, duration of admission - 6 weeks, course dose - 750 mg (6 tablets);
  • herpes infection (treatment): the first 2 days at 125 mg, then 48 hours later in the same dose, course dose - 1250-2500 mg (10-20 tablets).

When treating ARVI, including influenza, if signs of the disease persist for more than 4 days, you should consult a doctor.
