Pharmacological properties
Pharmacodynamics
Clotrimazole, the active ingredient of Candizol, is an antifungal agent used externally and intravaginally, belonging to the group of imidazole derivatives. It reduces the production of ergosterol, which is an integral part of the fungal cell membrane, which causes a change in the permeability of the plasmalemma and causes further cell lysis.
In small concentrations, the active substance exhibits a fungistatic effect, and in high concentrations (more than 20 μg / ml) it is fungicidal, affecting not only proliferating cells. When used in fungicidal concentrations, clotrimazole interacts with peroxidase and mitochondrial enzymes, resulting in an increase in the content of hydrogen peroxide to a toxic level, which in turn also contributes to the destruction of fungal cells.
The drug demonstrates activity against pathogenic dermatophytes (Epidermophyton floccosum, Trichophyton mentagrophytes, Trichophyton rubrum, Microsporum canis), mold and yeast fungi (the genus Rhodotorula, Pityrosporum orbiculare, the genus Candida, Torulopsis and Malaria versia versicolor) furfur).
Pharmacokinetics
With intravaginal use, the absorption of the active substance is 3-10% of the administered dose. High levels of the drug in vaginal secretions and low levels in the blood are noted for 48-72 hours. In the process of a fairly rapid biotransformation of clotrimazole in the liver, inactive metabolites are formed.
When applied externally, the drug is poorly absorbed through the skin and mucous membranes, and practically does not have a systemic effect. The concentration level of the drug in the epidermis is higher than in the subcutaneous tissue and dermis. In the deep layers of the epidermis, the content of clotrimazole exceeds the minimum inhibitory concentration for dermatophytes.
Indications for use
Vaginal tablets Candizol are recommended for adults and adolescents, after the onset of sexual activity, for the following diseases:
- genital superinfections due to microorganisms susceptible to clotrimazole;
- genital infections caused by yeast-like fungi of the genus Candida.

Candizol, instructions for use: method and dosage
Vaginal pills
Candizol tablets are used intravaginally.
Adults and adolescents are recommended to inject the drug as deep as possible into the vagina while lying on the back with slightly bent legs.
Recommended dosage regimen: 1 tablet daily in the evening before bedtime. The duration of treatment depends on the dosage: 100 mg tablets are taken for 6 days; 200 mg tablets - 3 days, 500 mg tablets - once.
After consulting a specialist, a second course is possible.
Cream for external use
Candizol cream is used externally.
For adults and children, the drug should be applied 2-3 times a day to previously cleaned and dried problem areas of the skin with a thin layer, then gently rub in.
The course of therapy depends on the severity of the lesion, the localization of the disease and the effectiveness of Candizol. Therapy for dermatomycosis is carried out for at least 28 days, for pityriasis versicolor - for at least 21 days. In case of fungal infections of the feet, it is recommended to continue treatment for at least 14 days after the symptoms of the disease have subsided.
Side effects
When using vaginal tablets, the following undesirable effects may appear: vaginal discharge, burning, itching and swelling of the vaginal mucosa, frequent urination, pain during intercourse, intercurrent cystitis, gastralgia, headache, skin allergic reactions, burning sensation in the penis in a sexual partner …
In the case of using the cream, there may be side effects such as tingling and burning at the sites of application of the drug, peeling and irritation of the skin, blisters, edema, erythema, allergic reactions in the form of itching, urticaria.
Overdose
There is practically no threat of an overdose of vaginal tablets, since the antifungal agent acts locally. There is no specific antidote. If an overdose is suspected, activated carbon is prescribed and symptomatic treatment is carried out, if necessary.

special instructions
In case of a simultaneous infection of the labia and adjacent areas (candidal vulvitis), it is recommended to additionally use Candizol in the form of a cream against the background of treatment with vaginal tablets.
In order to prevent urogenital reinfection, simultaneous therapy of sexual partners is necessary.
Candizol cream is not recommended to be applied to the skin around the eyes.
Do not treat areas of ulcers, erosions, wound surfaces with the drug, as well as areas with other violations of the integrity of the skin.
It is required to stop Candizol therapy if signs of irritation or hypersensitivity occur.

Application during pregnancy and lactation
In the first trimester of pregnancy, the use of Candizol is contraindicated.
In the II and III trimesters of pregnancy, as well as during breastfeeding, vaginal tablets can be used only if the expected benefit for the woman significantly exceeds the potential threat to the health of the fetus / child. In pregnant women, therapy with vaginal tablets should be performed without an applicator.
In the course of clinical and experimental studies during pregnancy or lactation, no negative effects of the drug in the form of a cream on the health of the mother or fetus / child have been revealed. However, the appropriateness of using the cream in pregnant and lactating women should be determined individually by the attending physician. It is contraindicated to apply the agent directly to the lactating breast.
Pediatric use
Candizol vaginal tablets are recommended for adolescents (after the onset of sexual activity) according to indications.
Cream treatment can be carried out in children according to the indications.
For violations of liver function
In the presence of liver failure, it is required to periodically monitor the functional state of the liver.
Drug interactions
With the combined use of vaginal tablets Candizol with other drugs, the following pharmacological interaction is possible:
- amphotericin B and other polyene antibiotics: vaginal administration of clotrimazole decreases the activity of these drugs;
- nystatin / natamycin: reduce the effect of clotrimazole.
When using Candizol in the form of a cream, negative interactions with other drugs / agents have not been established and are not expected, since clotrimazole has an extremely low absorption capacity.
