Azilekt is an antiparkinsonian drug; selective irreversible inhibitor of monoamine oxidase type B.

Release form and composition

Azilect is produced in the form of tablets: almost white or white, round, flat-cylindrical, with a bevel, engraving "GIL 1" on one side (10 pcs. In blisters, 1, 3 or 10 blisters in a cardboard box).

The composition of 1 tablet includes:

  • Active ingredient: rasagiline - 1 mg (rasagiline mesylate - 1.56 mg);
  • Auxiliary components: talc - 4 mg; mannitol - 159.24 mg; colloidal silicon dioxide - 1.2 mg; corn starch - 20 mg; stearic acid - 4 mg; pregelatinized corn starch 20 mg

Indications for use

Azilect is prescribed for the treatment of Parkinson's disease (as monotherapy or simultaneously with levodopa drugs).

Contraindications

Absolute:

  • Hepatic impairment of moderate to severe degree (classes B and C on the Child-Pugh scale);
  • Combined use with other monoamine oxidase inhibitors (including drugs and food supplements containing St. John's wort), pethidine. The interval between the end of rasagiline therapy and the appointment of these drugs should be at least 14 days;

Method of administration and dosage

Azilekt is taken orally, regardless of food intake.

The daily dose is 1 tablet (1 mg). The drug is used for monotherapy or as part of a complex treatment with levodopa.

Side effects

The development of serious adverse reactions arising from combined use with selective serotonin reuptake inhibitors, selective serotonin and norepinephrine reuptake inhibitors, tetracyclic / tricyclic antidepressants and monoamine oxidase inhibitors is well known. During the period of post-registration studies, there were reports of cases of the development of serotonin syndrome, which manifested itself as agitation, confusion, rigidity, fever and myoclonus, in patients who were simultaneously taking antidepressants / selective serotonin and norepinephrine reuptake inhibitors and rasagiline.

In the post-registration period, cases of increased blood pressure (including individual cases of hypertensive crises) were recorded in patients using an indefinite amount of tyramine-rich foods (cheeses, chocolate, etc.) in their diet, as well as when using the ophthalmic vasoconstrictor tetrahydrozoline.

There is evidence of cases of impulsive personality disorder in patients treated with dopamine receptor agonists and / or other dopaminomimetics. The same disorders were observed in the post-registration period when taking Azilect, in connection with which, during therapy, it is necessary to monitor the condition of the patients. Consideration should be given to the possibility of developing disorders such as increased libido, compulsive behavior, gambling addiction, obsessions, hypersexuality, impulsive behavior, and compulsive needs to buy or buy.

Combined use with sympathomimetics, dextromethorphan or complex cough medicines for oral or nasal administration containing ephedrine or pseudoephedrine is not recommended.

Disorders such as hallucinations and confusion can be a manifestation of the disease or be a side effect of Azilect.

Given the likelihood of developing significant adverse reactions from the central nervous system, during the period of therapy when driving vehicles and performing potentially hazardous activities, care must be taken until patients make sure that Azilekt does not have a negative effect on increasing concentration and speed of psychomotor reactions.

Drug interactions

With the combined use of Azilect with some drugs / substances, the following effects may be observed:

  • Pethidine: development of serious adverse reactions (the combination is contraindicated);
  • Other monoamine oxidase inhibitors (including drugs and food supplements containing St. John's wort): there is a high risk of severe hypertensive crisis (the combination is contraindicated);
  • Fluoxetine or fluvoxamine: the likelihood of developing an interaction (the combination is not recommended; the interval should be observed after discontinuation of fluoxetine or fluvoxamine - 5 weeks, after discontinuation of Azilect - 14 days);
  • Dextromethorphan, sympathomimetic drugs: potential for interactions (combination not recommended);
  • Potent inhibitors of the isoenzyme CYP1A2: a change in the plasma concentration of rasagiline, in smokers - a decrease (the combination requires caution);
  • Selective serotonin reuptake inhibitors, selective serotonin and norepinephrine reuptake inhibitors, tricyclic and tetracyclic antidepressants: development of serious adverse reactions (combination requires caution);
  • Entacapone: increased clearance of rasagiline.
